Hi I have just watched your informative video. I also noticed the time and effort you have put in to answering questions, that is so amazing! Much appreciated! I do have two questions if you or anyone else is able to answer: Can we make many more cultures from one syringe and how far have you been able to stretch a 10cc syringe? Is there any other way to propogate cordyceps for example from spores? Thank you kindy.
@SporenSprout Жыл бұрын
You can inoculate 5 quart jars of LC with one master syringe. That would make enough LC for 300 viable syringes or about 20 bulk bins. If you try to use some of that mycelium to make more LC, the culture will have less and less yield and more deformities the more you expand and according to your storage and time frame. You can stick a cordyceps fruiting body to the top of a water agar Petri dish with petroleum jelly and it will discharge the ascospores onto the water again and germinate. You can use a microscope to isolate single ascospores and attempt to mate them together and run fruiting trials to see if it’s viable, or you can select multi-ascospore colonies and run fruiting tests with that. I will be posting a video of how I collect the ascospores soon.

Well explained, have some questions... Once you got the fruits, how do you harvest them? Grain+Mycelium can be also be used? What is the best or prefered way to preserve them? Dehydrate?
@SporenSprout2 жыл бұрын
I just use clean scissors or a knife to harvest. I’d say to give the left over mycelium cakes to chickens for them to eat. Studies suggest that mycelium contains active compounds although I do not agree with those who process the rice in with the final product because it is just a filler. If you can separate the mycelium from the grains then it is better. Freeze drying is best because of the low temperature. Dehydration at lowest temperature works best.

@SporenSprout Жыл бұрын
Thanks! I’m actually gonna be posting a video about that soon. You can clone but the strain degrades fast and fruits become mutated and small so you have to collect ascospores and mate them together until you get another viable strain.

@SporenSprout Жыл бұрын
@@tswitch86 You stick the mature cordyceps to the underside of a water agar plate lid using petroleum jelly and the asco spores discharge onto the water agar and you can find them with a microscope and extract them from the plate one by one to individual plates. From there you mate them all together which takes ALOT of plates and work. I prefer letting the discharged ascospores stay on the water agar plate to grow and eventually all the different ascospores will collide with one another and in theory will mate with each other and dikaryotic mycelium will form on the leading edge. I then extract a section of the uniform growth suspected to be dikaryotic and transfer it to liquid culture to expand from there and test fruiting ability. You can verify dikaryotic mycelium by looking for “clamp connections” where two hyphae connect in the microscope but they are not easy to find.

Is there a way to transfer from agar to the rice or does it have to be LC? I've made LC of Cordyceps Militaris before but I only poured a little on sterile wild bird seed. I use that or rye. Only where the LC was poured on is where the mycelium grew.
@SporenSprout Жыл бұрын
LC is the only reliable way. Cordyceps mycelium rapidly degrades on agar due to oxidation. It is safe from oxidation while submerged in LC media. Rice is the preferred substrate.

Question: regarding 12:17 when you state “… to prevent the mycelium from replicating excessively” 1) Can you elaborate on this? Does have to do with the genetic particularities of cordyceps? 2)How old is this culture you are using here?
@SporenSprout Жыл бұрын
Yeah it is the genetic difference of cordyceps. The strain becomes senescent quickly when the mycelium is exposed to oxygen while replicating or when exposed to higher temps than 70F. That’s why it’s best to culture the mycelium submerged in liquid rather than on agar plates. The older the culture is the more mutations occur and less yield received. The culture in this video was around 1-2 months old, 2nd expansion from original syringe.
@hopehenley3002 Жыл бұрын
Thanks for these instructions. I have 2 questions that I still need clarification on. 1. How long do you inoculate the honey water in the dark? The average consensus seems to be about 3 days of darkness. What's your opinion? 2. After removing the jars from the dark (since you prepared multiple jars), I'm assuming that you need to swap out the jars on the magnetic stirrer (rather than CONTINUOUSLY STIRRING EACH JAR for a the FULL five days straight with each jar on its own separate designated stirrer). If you swap every jar to INTERMITTENTLY magnetically stir them, what is the estimated AMOUNT OF TIME they are stirred and HOW OFTEN during those 5 days? I apologize if my questions are elementary. Thanks for your patience.🙂

Growing in bins does not require a pressure cooker, only a flow hood and instant rice cooker. If you don’t have a flow hood you will need to grow in jars. You can try to steam boil the jars of rice for 2 hours instead of pressure cooking. The magnetic stir bar could be replaced with a piece of glass or marble but it could crack the jar or not break the dense mycelium up well enough.

Quick question: the red substance sealing the injection port - is that paraffin? I would like to make my own jars due to price reasons and noticed that you seemingly had a similar idea. Did it work? Do you still use the same ones or is it really worth it to buy the pre-built ones?

I really appreciate your effort for putting a very informative video on KZbin. Before asking I went through the comment section but no one has asked this question here yet to get my answer. What is the total lifespan of this LC in the syringe and what temperature they need to be storage throughout their lifecycle ? How do they ship them out to customers and how many days these LC can travel without getting spoiled. I am planning to take few syringes with me when I travel overseas that will take me at least 3 to 4 days. In this case should I look for locally or I can still take them safely with me from USA? Thank you again for your hard work.

The mycelium will generally stay viable for at least 6 months in the refrigerator. The mycelium should be stored in temperatures around 40 degrees F when not in use. The mycelium will be ok at 60-70 F degrees for your travels but it should be returned to cold temperatures quickly. It should not be expanded on agar and should only be expanded in liquid culture 1-2 times to prevent degradation.

Would LME instead of honey work for the liquid culture? If so, how much LME per how many mL of water? Much appreciated 👍
@SporenSprout Жыл бұрын
Yeah LME will work. I use a very tiny amount of LME (not even a full 1/4 teaspoon) to 600 ml of distilled water. I also add the same measurement of peptone as well.

My mind is a bit blown by the fact cordycepts degrades so quickly. Are there any other species you know of that do this? I’m a newbie in mycology but I am really familiar with fermentation. It reminds me of natto, which you can also only use the previous batch about twice before it’s contaminated/weakened too much to be edible a 3rd or 4th time.

Thank you so much for an super easy tuto for growing cordyceps, Can't wait to test it Do you think we could use honey instead of malt extract ? ( find of difficult to find that here ) And is nutritional yeast in powder and in flakes form the same ? Thank you again ! You rock
@SporenSprout2 жыл бұрын
You can probably use honey instead of LME. Yes both flakes and powder are the same.
@a110100 Жыл бұрын
I was going to attempt this for the same reason. Now I will with more confidence of replacing LME with honey.
