Plasmid Extraction from Positive Transformants
Plasmids are extrachromosomal DNA, which may contain antibiotic resistance genes. In recombinant DNA technology, plasmids are used as vectors to clone a gene of interest which may express or produce proteins with biotechnological applications. Plasmids are usually transformed into Escherichia coli competent cells grown in LB broth media amended with appropriate antibiotics. Positive transformants expressing the antibiotic resistance genes in the plasmid can thus be easily screened from the selective medium. Conventional method of plasmid extraction uses the alkaline lysis method.
