Ya...I used alcohol, and lit my lighting inside, and my box was air sealed with gloves.....it exploded. It was an awesome mad scientist experience. Lessons learned.

Lesson learned the hard way myself. 2nd degree burns on my neck and my hair is scorched to the scalp in some places. Bleach works fine, and alcohol on towel inside if any. Wipe items before putting in the box.

lolll glad I'm not the only one, I just flashed my box like 2 or 3 weeks ago, my right hand is messed up but it healed now. Well, at least the box is extra sterilized from that flash. I actually thinking of a safer way to flash my box like that again because in my theory the flash will push any particles out and the heat will destroy any microbes inside.

Really appreciate you going through these techniques w/out using your FH. I'm surprised how many YT channels go though basic techniques (like inoculating a bag) sitting in front of a flow hood. I always refrain from working directly in the lid as most of the dirty particles like to chill there. I use a small shelf divider to work an inch or two above the lid

I'm going to try using a damp towel soaked in water with ~5% bleach to seal the floor of my SAB and just leave the lid off completely. I saw MycTyson using this method.

For SAB holes if you take a 28oz food can and put it upside down on stove for a minute, and then in a well ventilated area using a glove use that to melt your holes. Clean melted plastic off of the can in between doing holes, works perfect!

I just got my sab and wiped it all down alcohol 70 same for the out side of the all in one grow bag put it an my spore syringe in I had a new in the sealed package needle so I didn't burn it hope that was not a mistake and it set it up all in my closet so it can stay in the dark and at a 70 degrees I picked a strain called butters but now I don't have a clue what to do next

Shouldn't be hard at all to make clean agar. Maybe it's where you're storing it that is making it dirty. Look at no-pour teks if you still have trouble, but put your agar in new (already sterile) ziplocks or even just spray the insides of already opened ones with alcohol or Lysol before putting in the plates.

I have wanted to do this for some time. Just a thought, Sodium hypochlorite is a great disinfectant you can find it in Pool cleaning areas Lab content would be 80% but the pool stuff is great for many things. Used to clean well water and food products in major industrial areas. Also a main ingredient in killing fungus growth in wood framing such as under houses and basements... .. . (Perhaps not needed). Something more, I use a similar box for jewelry work to capture dust I keep the box right side up and silicone a picture frame glass to the top so you can see better.

Spore syringe is easily acquired. Liquid culture is normally made from an existing fruiting body through a process that can be found on KZbin. Liquid culture is a nutrient rich suspension medium while a spore syringe is just sterile liquid that spores float in until use...from what I have observed. I started with spore syringes simply because it was the "novice starting point". With more research I moved to liquid culture and agar transfer.

I put pvc rings on my SAB that zip lock bags fit super tight on that act as sleeves so just my gloved hands and bags go in and it sits on a stainless steel surface. The bags are probably overkill but it sounded good at the time

That's absolutely possible! Mycelium expansion increases exponentially. You could then turn that into thousands of pounds of fresh mushrooms given enough space and resources.

he said in the video to use a 50% bleach/water solution (1:1 ratio); I highly recommend NOT doing that as it's noxious. 1:10 bleach to water ratio is sufficient. You can google around for bleach concentrations needed for sanitizing. They'll often use even less than 1:10.

ideally the cut sides of the parafilm never come close to the opening of the containers... They stay closed, you can contaminate the outside surface all you want, the parafilm keeps it closed anyway. Never had a problem of contamination going across or through parafilm if that's what you're worried about

Correct! They are typically gamma irradiated by the manufacturer. That’s why nurses don’t flame sterilize before giving you a shot. 🙂

How long after sealing them would you inoculate it? The residual moisture left when it cools after shaking leaves grains wet. Would you let the bags sit a few days or use them.

I’ve never had that be an issue. The moisture is drawn into the grain in about a day but I use them as soon as they cool down.

That’s a very valid question. In the open air there’s still air currents that carry spores and things around. Inside the still air box we try to let things settle and hopefully keep contaminants at a minimum.

Great video. The main reason I haven't jumped into a still air box is that I don't want to do the nuclear method of 4" PVC into thick rubber gloves. I like your method of just sliding clean hands through the ports. Do you find that this still gives a pretty good success rate?

Actually, a lot of people unknowingly set themselves up for failure installing thick gloves into the box. Gloves create a billows effect that pulls in air from around the lid of the SAB. So when you move your arms around with the gloves installed you’ll literally pulling in dirty air from outside the box. Having no gloves avoids this all together.

You heard right and he is a bit off on a couple things. f you look at the label on any cleaner it will give you the optimal ratios. If someone does not believe the label, ask yourself this...why would they lower a ratio when increasing the raito would have you using more product and earning them more revenue. It makes no sense. Generally they will give you the best mix for the job at hand, One change to his video would be to let the bleach mixture dry. For the most part, none of these compounds kill on contact, they need time to penetrate the organism to kill it. Just like alcohol, you need to let the material sit for a bit before you wipe it down...if you do at all. In SAB work and when it comes to contam, slowing down a bit will go a long way.

A flow hood is much better and you can likely use alcohol in it. The reason alcohol is bad in a SAB is the alcohol evaporates and sits in the box. Add a flame and BOOM! Well a gush at least. In a flow hood the vapor is pushed out.

Bleach takes time to work. Alcohol is fast acting. They work completely differently. Ideally bleach first, let dry, then alcohol to sterilize as you go. But it all works if you stay clean as you said.

How long does an inoculated agar dish live?

It can take about 2-6 weeks for the fungus to grow out on the plate. If there are no signs of contamination I'll move them to the fridge. They can easily last 6 months in there. The only real issue is the agar drying out.

@@nikkifigueroa4441 if you just growing for fun here and there you don’t necessarily need one, the still air box a great idea but if you have a sterile work area with little air movement and take the precautions needed should be able to grow some mushrooms

It's literally just blue food coloring. You can use any color you'd like.

Why 50/50 bleach? Isn't a 10-to-1 mixture sufficient?

It's honestly just personal preference. 10:1 works fine. Just test and see what works for you.

1 : 32 ratio is effective, that's 2 tablespoons per quart (a quart is 64 tablespoons) for disinfectant use. I like to go with 5-10% which would be about 1 : 16 or 4 tablespoons per quart.

I’ve been considering making one of these . Until I have the money to buy it make one , does a small Hepa air filter work? And also do you keep the grain in the still air box until it’s fully colonized ?

It would be best to keep the bag in a dark and warm space. You want to find out what temperature is best for that particular mushroom but it's also pretty easy to create your own incubation chamber that allows you to control the temperature.

Great video thank you. I watch a lot of these videos and I enjoy your style. 🙏 Question, with 1/10ml syringe of liquid culture how many 5 pound bags of grain spawn can I inoculate? Some people say two bags so 10lbs of grain spawn and other people say they use very little and find great success with only using 2ml per 5lbs of grain spawn. So the 1/10ml liquid culture gives them 5/5lbs of grain spawn. Now that seems like too little to me and my next question was did those bags take forever to fully colonize? Or was it similar in time to how long day 2 bags so 5ml in each 5lbs bag took to fully colonize. But they never got back to me with the answer :(I understand people are busy and I just appreciate any advise I was given!:). So I was wondering what your take is? Also if I may ask another question, how much of a ready agar plate would you cut up and put into grain spawn? I know to use the nice middle sections that look the best, but to me I feel like I could use each agar/Petri dish for a few, maybe 2-4 bags 5lbs bags per Petri dish. Another move to add a layer of protection is to take a pair of large, rubber kitchen gloves, glue the rim of them around the hole and cut out the hand portion so you can stick your hand through, this way it will reduce the airflow into the box while you are working while not inhibiting your hands as much as in a traditional glove box

Adding gloves actually causes a billow effect and will stuck in air around the lid and possibly pull in contaminants.

@@midnightmushroomco.2091 how about a tyvek filtered port to ensure no vacuum possible. Seem like a good move? (I’ve never done this, I’m just on my like 8th SAB video lol, 9 more and maybe I’ll make of these)
