After watching your videos for the past month I have successfully created my own: SAB Spore syringes (totally sterile) Agar plates done with your solo cup idea Growing spores on agar (totally sterile) Isolating my first genetics Successfully developed my own unique recipe of grain I have a shelf full of about 30 jars with mycelium running like Bolt Spawned three shoebox tubs and it all looks great. One main critique: Do ALL your shaking OUTSIDE of the box. Get your primary spawn jar broken up then place into box. Once you add the spawn to the next jars, just tighten the lid and set aside. As always, keep movement to a minimum inside the SAB. Once you have done all the jars, pull them from the SAB and spread that spawn. One thing I love about starting this hobby is there are hundreds of way to go about things. I am always learning and trying to dial in my own tek. Thanks again for another great video!

It's crazy how your 90% colonized jar looks so much less colonized after the shake-shake.

That's because technically it was only 20% colonized if that. A fully colonized jar of grain cannot be shaken loose, you have to go at it with a spoon (sterile) and work at scooping it out. If you can shake it loose and see grain, it's not ready yet for G2G and certainly not ready for spawn to bulk.

iv found its worth adding a few layers of 1-3mm vermiculite in LC grain jars if you dont shake during colonization you end up with so many tiny inoculation points when you use it for g2g.

Always love the videos and the quality. Just one question -- is the wire cooling rack you always use in your SAB actually doing anything? Assuming you wipe down the bottom of your SAB as well as the sides when you begin, it seems like it does nothing except introduce instability to whatever you set on it...?

it provides an elevated surface to work on, and in my case it does provide stability because im using the lid of the SAB as the base and its flimsy and not really a true flat surface.

Hey PGT, have you had any increase in contam rates with those unmodded lids? How do you take them out of PC - Do you wait for them to cool before opening PC so contamned air can't get sucked in before you screw the top? I made some screw top no pours and was stunned when they triched out in the few minutes they were a quarter turn loose when I pulled from PC.

Great, my friend just did a g2g and its working! Do you count it as a 2nd Generation Mycelium because of its Maturaty. And is it a good way to Isolate a Specific Strain? This is my friends new fav hobby, he said it's quite easy once you know what your doing. Hes on his first bulk grow, thx for knowledge

At this point do you leave the jars cracked a little to allow some oxygen in, i have watched most of your videos and i just found u and subbed, i think u said in a earlier video to keep open lid a liitle like a quarter turn or something but i dont remember for sure, as i have a bad memeroy. Sensei! I love your vids this is my first time growing I have one question though, I inoculated two bags on the same day and kept them next to each other and one of them is doing so good already spreading mycelium and the other one looks like is doing some sort of oxygen exchange and its holding a stable humidity but I'm not seeing anything at all. Is this normal?

Newbie mushroom grower here. I have played around a bit with those mushroom blocks you buy to grow mushrooms. I was wondering if anyone has experimented with using those blocks after one or two fruitings, as an inoculant for another batch of mushrooms. I was thinking of using a 5 gallon bucket, with holes, of fresh substrate and an old "log" crumbled up for an inoculant. Anyone tried this, and if so, how did it work?

I think the mycellium bruises and changes color when it is broken up and turns white again after a few days. Maybe PhillyGoldenTeacher can elaborate

​@@spewdog Nah it's just because it's so fluffy, like cotton candy. A thick fully colonized agar plate will become just a piece of material around the size of a grain of rice or smaller on your finger if you swipe your finger around the whole thing. But it leaves small amounts of mycelium everywhere it already was before the shake and after it had already eaten from the grain so it regenerates easier.

@@VladTheImpalerTepesIII That's not true though. Mycelium grows outward every direction if it isn't on a 2d surface like agar. If the jar looks fully colonized from the outside of the jar, it's good on the inside too, for a transfer at least. A jar doesn't need to be consolidated so long that it refuses to break up, to be considered fully colonized. Neither me or anyone I know, including professional growers, let it get to that point before G2G. Although I do let them sit a few days or so after fully colonizing, to consolidate, before spawning. There's a difference between the two. Colonize just means when it's covered the grain, consolidate is when it has more time to digest it, and it gets thicker during this time. Also pouring grain for G2G is better than a spoon, so shaking it is something you should be able to do. But if you're not waiting a while for your jars to consolidate like that, and they're still hard to break up, it could be a sign of bacteria, which normally will still fruit, but not to it's potential.

Is there a way to extend the shelf life of a “donator” jar of inoculated grain? Such as in a way where this donor material can sit for months and be used from time to time without the mycelium expiring from lack of nutrition. Can this be achieved by a fridge? Or adding food from time to time? Thanks for any thoughts on this

My question is that I'm about to transfer a Uncle Bens Rice bag to 7 Qt. jars of Oats. My Oats expanded a lot more than I was hoping. Would it be a bad idea to spoon out a couple inches of the oats before I add the rice? I am working in a SAB.

I have UB bag that has colonized grain on top that I want to use for g2g. assuming i use a SAB and sanitize properly - can I transfer uncolonized rice from a freshly opened UB bag to a grain jar with GE lid and spawn from jar instead of using the manufacturers bag?

any clean jar will work. he keeps the jars loose a quarter turn for air :-)

Would grain to grain transfer work if your colonized jar is essentially 90% colonized or is too far along to do so? I have an extra jar that wasn't ready at the time I moved the other jars to the sub. it has solid myc growth covering the entire jar and only a few grains are left to colonize

It's an unmodified method that leaves the lids a quarter turn loose for exchange. It's mentioned in 'The ultimate lid tek guide' video 🙏🍄

2 options: plastic box with 2 glove holes (cheap option with low space) or flow hood with H14 filter. You can try your luck soon after shower in a toilet without carpets that u just sanitized like a OCD mom spraying alcohol on all the walls with all the windows and doors of the house closed using a mouth mask. Out of those 3 options its just too risky to get contaminations.. Like.. Those spores are literally EVERYWHERE.

PGT, Do you have any ideas of how I might maintain the room temperature once my substrate & grain go into my monotubs? My home sits at about 16 degrees Celsius (60F) throughout most of the day and night. Will that negatively affect my growth? Or just slow it down a little?

You can for a few rounds, but not indefinitely. It will reach a point of senescence.

When you are having your grain colonize in jars, do you need to have any air exchange holes with polyfill or filter patches? Or can it stay completely sealed. I know a self seal injection port is good for liquid culture for injections.

You can technically get by without any holes or modifications on the lid but growth may/can stall. I would leave the lids loosened 1/4th turn and flipped upside down if you aren’t using any holes or mods. If you use mods then use the micropore d surgical paper tape or polyfill

looks to me every jar is hermetically shut hence no air exchange of any kind. mycelium growth does not need oxygen to happen?

This is why I think sealable bags are better than jars for your mycelium growth. It seems to make it alot more convenient to be removed when it's ready

What if your grain is fully colonised and the myc is to strong to break and shake, how would you do G2G then?

Can you keep doing grain to grain indefinitely? Thinking of it as sort of like a sourdough starter where you can continuously add to it and multiply it

Technically you can but it makes sense to start from time to time from agar to makes sure that your culture is not contaminated.
