Duuuude!!! Just struck GOLD finding this video!!! Been struggling with bacterial riders for a whiiiile now. Basically have narrowed it down to my LFH filter been saturated, and every "open jar" agar/grain transfer I do in front of it gets bacteria that I later transfer to tubs and get Trich. Really frustrating cuz the myc looks "OK" but at the end of the cycle, all that effort and resources are lost. But, looking at your video I've learned that they may ALSO be embedded bacteria coming in the genetics purchased. 🤔 Cheers man! Keep doing these videos! One about possible origins of such contams/bacteria would be nice!!!

@@Mycophiliahe means embedded bacteria. Basically youd leave 1/3 of the dish with the bacteria embedded mycellium then empty the rest out and fill with fresh agar alongside that wedge and then cut a "ditch" out of the agar so there is a line of clear space between what's already colonized with the bacterial mycelium and the side with the fresh agar. The mycelium will walk over that gap but the bacteria in theory wouldn't transfer along with it. I haven't personally had to do this, but it certainly does make a lot of sense

One idea I wanna share with you @Mycophilia: I remember Mycoplasma are smaller then other bacteria and prefer to stay intracellular, not intercellular. Very interesting, but what makes you think the jars with the white mycelium are contaminated with bacteria, or even contaminated at all? Have you done some tests that support that notion? Might not something else actually be going on? (BTW, next time why don't you slice the white mycelial mass into 1.5-2 cm 'breadslices' with a knife and try putting them btw a thin 50-50 CC-Verm bottom and thicker 50-50 CC-verm top layer and see what happens. I suspect breaking up spawn that looks like that might not work very well.)
@Mycophilia Жыл бұрын
They are 💯 contaminated, once you’ve grown for some time you know what’s contaminated and what’s not. I have listed a number of reasons on this video, the primary ones being the extremely thick growth that makes it impossible to break up, along with all previous attempts with this culture always ending up with the same green mold, at least those that have been spawned to bulk. The ones that have not (bottle fruited), have not fruited in 2+ months of fruiting conditions, in addition to growing very strange mycelium masses after the first month, a hallmark of contamination. As for your idea of slicing, a better method exists called jar/top fruiting. It is better because you don’t need to break up the mycelium at all. Hope that helps.

Question, were those white jars of the compressed myc started from what looked like a singular culture on an agar plate from spores? I think people get germinating spores mixed up at times. Spores can germinate and grow by themselves. They only need to meet up later with other germinated spores to go on and grow healthy mycelium and eventually fruit. This may be the problem with that myc. I'd try introducing another sample of myc that came from the same strain and see of they marry 🤷

Üblicher Weise fertigt man aus dem Petrischalenansatz erst einmal Flüssigmycel an. Dadurch lassen sich viele Probleme mit Kontaminationen vermeiden.

I think I have the same problem. Transfer through antibiotic agar could help with those embedded bacteria (gentamicin, chloramfenicol)?
@Mycophilia Жыл бұрын
It should, I would recommend doing 2-3 xfers on antibiotic bacteria, then 2 transfers on normal agar to make sure it doesn’t come back (as bacteria will always overcome the antibiotics eventually, so you need to make sure they’re gone for sure before putting it to grain)

So what if your spawn bag had some wet rot maybe not wet rot but a few slimy spots in the beginning but then the mycelium still took over and properly spawned? Is it still good to use? I went ahead and put it in a montotub and it looks like it’s colonizing properly
@azn4109 Жыл бұрын
Let me say it better I did too much syringe and it was a little wet took a few weeks to colonize but it did so is it still safe * newbie
@Mycophilia Жыл бұрын
It’s not an automatic death sentence, fruits aren’t guaranteed but with wet rot you can oftentimes get a flush or two before trich takes over.

Those "embedded bacterial" jars are not actually contaminated. I think he's mistaking crappy genetics. Those don't look bad at all. There's no "tell tale" sign of anything in those jars

would all of those take over the myco a month after innoculating mss or is it always much faster

I would never thought that unbreakable mycelium is contaminated...I am no expert tho.... maybe anti bacterial agar would help... make clean coulture...?

Why one would bother with PF tek anymore is beyond me. But each to there own. First I had to pressure cook the jars, drill holes then squirt 2ml of liquid spawn into each jar. I did 4 jars, lost two cakes to bacteria, then put the other two in a tub with perlite on foil. (It took a month to colonise the jars btw). One cake produced a few fruits then that was it. I’ll never do it again.

Yes, I have a video on cleaning contammed jars

Apologies if you've already addressed this in a previous video, but when fruiting truffle producers, will the fruits be more potent the longer i let the spawn colonize? Or as soon as the jars is colonized(little to no truffles forming) spawning to fruit will be the same potency?
@MattRoadhouse Жыл бұрын
the fruits are tiny and potent as it is. Recent run was 6 months truffle then fruiting trays with peatmoss casing, but I'm going straight fruiting for Tampa / Mexi this round. Will see if they are in the same ballpark.

Thanks for the good oil! i have a similar situation with a H.PES genetic... i was sure its carrying something, but i cant see it on agar. there is a new tek available that is being tested in the community btw. Rit dye agar.... the other option would perhaps be water agar ?

In regards to the bacterial spawn, my grow bag has looked like that since inoculation. I was informed it was “tomentose mycelium”. It’s grown fine and now It’s in its 3rd week of fruiting conditions, not much has changed. I’m starting to get concerned that it may never fruit. What are your thoughts?
@Mycophilia6 ай бұрын
3 weeks and no sign of fruit is very slow. From my experience, I would definitely bet on bacteria being the case.

In a shoe box or tub can you have to much spawn and not enough sun will it give you a very small flush ?
@Mycophilia4 күн бұрын
Nothing to do with sun but too much spawn and too little substrate will result in reduced flushing performance.

I'm on my first attempt, and at first, the mycelium was growing beautifully, but today, I noticed a spot of localised green mould near the top of the jar. What would be your advice? It hasn't spread yet, and it would be easy enough to scoop it out with a sterilised spoon and discard, but I'm worried that the whole lot might need to be thrown out.

Could iodine be an option used for sterilization of equipment?
@Mycophilia Жыл бұрын
Haven’t personally heard of it being used for that purpose in mycology but I could be wrong. I just wipe things down in alcohol and flame sterilize equipment that will directly be touching cultures like inoculation loops and scalpel blades
