Loving this! I am still learning, love your lectures, they are so nice and clear. I wanted to ask about cells in suspension. For my cell type (lymphocytes) I know that people adhere them and I was just wondering similarly to what you say how much this affects the readout. I gues I would just have to compare the values. This was very useful, also the experiments with pyruvate with and without oligomycin for total respiratory capacity. Extremely useful! Thank you so much!

You can measure oxygen consumption in suspensions with an electrode system like an Oroboros respirometer or any other Clark-type electrode system.

Dear Prof. Kowaltovski, all your lectures are brilliant! Thank you very much for them! May I ask a question about the scheme for XF measurements from Bioenergetics book? According to the scheme, pH is increased when the sensor approaches to the cells. However, as far as I understand, it should be the opposite: if glycolysis occurs the acidification of extracellular medium will take place, thus pH should decrease. Could you please explain if I am mistaken? Thank you!

Thanks! Not sure I know what scheme you mean, but probably you are looking at ECAR - extracellular acidification rate, not pH.
