The heat shock is happening through CaCl2, which is counteracting the electrostatic repulsion between plasmid DNA the bacterial cellular membrane ( liposaccharide layer), mostly E.Coli as it is most competent bacteriathan for these procedures.

Watched this video to understand what the term "DNA Cloning" means and how that's done with the help of recombinant DNA technology. Got to know that it's reproducing of the desirable genes/sections of DNA with the help of certain restriction enzymes, DNA ligase, and the host bacteria that supplies plasmids to attach the target genes and then reproduces them. This video also clarifies the question - "How to engineer microorganisms?" - for which I have seen other videos earlier (insulin production as a case in point)

I have a doubt sir. Like u will collect DNA fragment of a virus r plasmid of a higher organism then u will maintain it in such a organism within which u can easily insert a foreign DNa fragment Then which DNA acts as vector here N how many organisms DNA will be involved ..??

How do we know for sure that the bacteria will express the genetic information from the plasmid, for example, even if the colony has taken up the plasmid, why would we be sure that antibiotic resistamce will be expressed?

I was wondering the following, this video tells me that DNA is taken from one organism and put into another organism's plasmid so that that organism can go and create a desired protein. My question is then, does the bacteria know how to splice the introns and exons when it's creating the desired protein. Much more logical to me would be that they take ripe mRNA, (already spliced correctly) reverse transcript that and put it in the plasmid, since bacteria don't have introns and exons.

Isn't bacteria a prokaryote and prokaryotes don't have introns.

Because we are using plasmids which carry the factors coding for resistance against some antibiotics and the scientists who perform these experiments are well aware of the plasmids and the resistance they have ..and as the replication occurs in the E coli this gets amplified...

Can someone answer me PLEASE!!! About how do we select the bacteria that took up the plasmid .. what I know that the Plasmid has antibiotic resistance gene NATURALLY (because he said we place it!), so obviously the other bacterias in the plate also have it, so they won’t die ?

the whole idea behind the plating and growth is to allow the bacteria with the resistance to grow and then bacteria that don't have the resistance won't grow colonies. Now that being said, it is science and sometimes the other bacteria will also grow due to naturally gaining the resistance, but that doesn't mean their numbers will be profound.

you can typically run a gel electrophoresis of ur isolated gene of interest next to a gene ladder that will show the expected bands of that particular gene of interest. If the two sets of bands correspond with each other, you know you have your correct gene of interest!

You would just add a restriction digest that flanks either side of the gene of interest.

closer but still not able to explain completely....how will we all of sudden will introduce antibiotic resistance gene in vector...won't it too will take process of colony formation selection of transformant same as that of our gene of interest...so by your video concept before clearing give rise to another doubt

How do you know if the plasmids taken up by the bacteria also contained the gene of interest? Couldn't some of the bacteria take up plasmids that gave them antibiotic resistance but did not include the gene of interest?

Because all plastids contained both antibiotic resistance and the gene. So either both went in or none
