I really want to understand how to prepare the inoculum. I still don't know if I need to dilute the inoculum from 10ˆ8 until 10ˆ5, it means 3x diluting it 1/10 or something else.

Thank you for the informative video. That would be great if you help me with the confusing part on 3:55 which is animated by transferring 800 micro liter while the mentioned transferred amount is 100 micro liter, besides considering the dilution calculation formulas the proper amount to be transferred from stock solution to make the 4 micro-gram / ml solution should be 0.08 ml or 80 micro liter.

It might be possible that there is a calculation mistake in the video... I dont really remember how i made the video. My suggeation is go by the formula

I have a question at 5:00, if add 50ul of the antibiotic at 4 ug/ml to 50 ml agar, does that change the concentration of antibiotic?

Bro one more doubt. What is the difference between the red coloured solution and gray coloured solution at time 5:04. Because both of the solution is labeled with the same ' 4ug/ml antibiotic sol' .
