Fwiw - I have MEcfs and am a responder to rapamycin, an mToR inhibitor -- it has increased my exertion ability and when I do get PEM, it is less intense and shorter duration. I initially took 1mg daily - it look less then a month to notice effects - currently take 1mg 3x week.

How long have you been on it? Improvements increasing over time?

Thanks for uploading this, the findings are very interesting, this could have some legs to it.

1 min on the treadmill would have had the same results. Pushing mice for 15 mins is normal even for wild mice.

I have severe bedridden ME/CFS and I know what severe suffering is like. So hearing you are inflicting it on innocent mice is disturbing. Animals aren’t objects. They are sentient conscious beings that will suffer too. Over 90% of animal models for drug research fail before human trials. Please use the mini organoids instead, you can even link multiple mini organoids representing different tissues and organs together. Stress and test that system instead. As no animal testing and suffering needed

Organoids would be completely insufficient for this test and make it way more complex. They would then advance to animal trials either way. To use organoids we would have to uncover many more pathways linked to CFS to then garner results. What you're suggesting would add many years onto the research.

single organoids, but not grouped system organoids

I had severe ME/CFS and I would sacrifice thousands of mice, thousands of monkeys and thousands of dogs for the chance to get little better!!!

@@kmkvladne Thats because people often see animals as objects but forget there is a living conscious being inside them, plus not realise the amount of suffering inflicted. But if your could feel the actual animals suffering, like if their pain, sickness and fear was directly linked to your brain so you felt it exactly what they do, you would beg for it to stop because it's unbearable suffering, as is far more intense in labs using animals as often no sedation is given. I used to work in 2 labs that tested on animals, and its horrific what they did to them. One evil standard test still done today is the LD50, Where they poison all the animals by increasing the dose up higher and higher. Sometimes over 100 times the therapeutic dose. 50% of the animals eventually die after days of agony (LD50 means Lethal Dose 50%), shaking, convulsing, vomiting, and screaming. This is to establish a toxicity level, that often and usually doesn't even match toxicity in humans. Other animal tests nearly caused certain medicines to be removed from ever being released. Paracetamol for example causes birth defects in dogs. Another issue is that 98% of drugs tested on animals fail to even work in humans. Thats millions of animals suffering by being poisoned to death for nothing. I have severe ME/CFS and the suffering is bad, but I'm not a coward that wants to make innocent souls suffer for me. Thats cheating. because theres plenty of other alternative testing platforms that exist and others that are yet to be invented, and the reason those technologies have been delayed until recently is because scientists lazily used animals like machines. >>>"Necessity is the mother of invention"
