So here's a tip for drilling holes in plastic. Start the hole, but just let the center cut in. Then, switch the drill to REVERSE. Then drill the hole. The drill bit will create friction, and melt the hole to a perfect circle, and there won't be jagged edges, and there is less of a chance of the hole cracking from the drill.

note that you cannot have the uv light on while working in the box or it will damage your eyes and microbes require at least 15 minutes exposure to kill them. "Long-term exposure to even small amounts of UV radiation can increase your risk of developing a cataract or macular degeneration. UV exposure adds up over time. Long-term UV exposure can also causes tissue elevations on the surface of your eye. These are called pingueculae and pterygia. Using sunglasses faithfully when outdoors may limit these." Also, I would recommend at least a triple layer MERV 16 material.

The plastic should be enough to block the UV-c rays. Granted you don't want to spend a lot of time with your hands in the box, but if you multiply time spent by 10, so 15 minutes times 10, assume 150 minutes of uv exposure. How many of you would think twice about being in sunlight for 150 minutes? Wear sunscreen if you're really worried. Or you know, long sleeves and rubber gloves. UV lights can be very dangerous if used improperly, but a few VERY simple steps make them just another tool.

OK I have two points to help out all the people below having problems with 1. not enough pressure to push air through the filter and 2. the people who are concerned about air moving in the box. 1. Its a bit of work, but if you disassemble the A/C infinity fan unit and turn the individual fans around inside the unit. the fan will now be sucking air through the filter instead of trying to blow it through. The air wont "bounce" off the filter. now install the fans INSIDE the box so its sucks air through the grill which is now outside the box and is sucking air through the filter into the box (still positive pressure in the box) It takes up a bit of space in the box, i know. but it works better. 2. The reason we want STILL air in a SAB is because the air isn't filtered, Its full of particles. we want the particles to settle out of the air and on the surfaces of the box, so it does not land on our culture/spawn/agar. but once the air is filtered and there are longer any particles in the air. it can move around all it wants. because there are no longer any particles that can settle on our culture/spawn/agar. I hope this helps a little

The fan should be mounted so as to create positive pressure within the SAB. In other words, the fan should be blowing air into the box. The filter should be a HEPA filter which will trap particulates down to 0.3 microns and keep them from entering the box. The positive pressure will keep particulates from entering the box from all the holes that have been drilled in it. I personally would not drill the small holes in the sides. The holes for your arms should be enough. If you are making a gloved box then you need the holes in the side.
@TOPskunkfisher2 жыл бұрын
Did you watch the video it is setup to produce positive pressure, this is a killer budget flow tote not a professional laminar flow hood, clearly not the same but definitely better than a still air box or nothing at all and it's a minimum investment

Built this, I accidently used a merv 17 filter but it's too dense for the fan to push air through. I also realized immediately after building this, that the science doesn't really work out. Sterile technique in a flow hood is to keep everything 'upwind', because contams can be blown off anything upwind onto whatever is downwind. In this design, your work is constantly downwind since the air is traveling across. The original design had the right idea with the fan blowing towards you. Like others have said, open agar plates would be the best way to test in a real world scenario, a meter won't be able to accurately account for an actual working environment where your work/hand is always upwind blowing off contams and turbulating turbulent air (because it's not laminar) even more

Hello guys. I'm new to mycology, just did my first grain preparation, autoclave sterilisation, inoculation about a week ago. Needless to say there is a fair bit of contamination but I'm glad I got that out of the way and I'm ready to do it again, this time on agar in a still air box. I have a chance of buying a 2in1 beurer EPA purifier/humidifier (110$) and I'm fiddling with the concept of whether running it for an hour or so in a chosen innoculation room coupled with a simple still air box setup would work. The model of purifier seems not to come with HEPA filter but rather EPA filter, even though the small research I've done indicates mold spores are relatively big (4 microns). Is this setup better/cheaper than building a laminar flow hood, considering the machine also has automation functionalities, which seems like a perk when it comes to fruiting. Thanks anyone
@c.odubhlaoich29482 жыл бұрын
Cheaper, yes. But a legit flowhood really can't be beat. A lot of people would also probably tell you don't even worry about that set up and just make sure you have sterile procedure down with a simple SAB though. Get a towel wet and ringed out with 1:10 bleach/water, lay it out flat and put your SAB over top of that and have some kind of cookie sheet or something to elevate your tools and jars off the towel, and spray the insides of the SAB with soapy water and you're good to go. Anything more than that is really just peace of mind, so flowhoods are really nice just because you can work openly and know youre doing it in a clean spot, but plenty of experts know that a simple SAB can be very sterile.

@johnstamos46292 жыл бұрын
yea.. it doesn't make sense to filter air on the way OUT. LOL
@ryanpongracz80513 жыл бұрын
Also your description of this being a negative air pressure system is false. This is positive because the intake flow is higher than the out take flow. In order for a flow to be negative there would have to be a seal to the environment to start, and the air intake would have to be lower than the outtake therefore making a suction effect in the intake flow and creates a slight vacuum to the room, thus making it negative pressure. This is definitely positive pressure.

You would think so. I built one and installed MERV 17 filter medium. Why not? right?The fans are too weak to blow through it at all. So I ordered MERV13 and the flow you get is adequate to pressurize your box if your vent holes arent too big or too many. These fans are not strong. There is actually more air blowing out the intake as a result of back pressure caused by the MERV13. Doubling the MERV13 would probably stop the air flow altogether like the 17. You need adequate pressure to be able to keep particles from entering the holes. That being said MERV 13 seems to allow adequate pressure at the vent holes with my arms in the arm holes. How well it filters remains to be seen. I'll report back.

Willy my boy youve got a lot of great content. Tons of valuable information, tips & tricks! I gotta tell ya bro you really screwed the pooch on this one! I followed your direction to a T! Tryin to push air through that filter material with those tiny little fans is like tryin to blow a fart out your ear! Fans could barely move air to the other end of the box without the filter? The UV-C doesn’t have high enough nano to do anything except piss off a germ! You lost ALOT of cred on this! It’s a great theory but you need to sharpen your pencil on this one! Much Love!

Good info but I'd love to see more of what's being talked about, less of the speaker. I learn better by seeing as well as hearing.

Where does one get the particular measure device?
@omma-llama78603 жыл бұрын
It's called an air quality monitor, detector, analyser, etc.., you can find them on Amazon. If you want the professional level indoor air analyzers, you can get those from Grainger.

Why is the position of your filter catching the debris IN your box? Why not attach the filter to the outside and catch it in the filter before the air enters? That way when changing out it's also less likely to get anything in the box.
@ryanpongracz80513 жыл бұрын
My solution was to put one pre filter on intake side, and two stacked on outtake side. Encased fan in filters to stop contams. Will kill fan faster than intended, but who cares, they are cheap and this works way better this way.

@@hughjasole673 nice. Mine seems to be working fine. I kinda prefer the positive air chamber because air is always blowing out the arm holes so nothing can get in
