This is a revisit of when I make benzocaine like a year ago. I was appalled at how unenthusiastic I was and how sloppy my video was.
The next thing that I put my money to is a proper vacuum pump. My current one smokes after running for 30 seconds. When I get a better pump, I will be able to do some proper vacuum distillations.

lol i read a story in my local paper about some drug dealer getting ripped off some guy managed to sell him a breeze block taped up with an hole in the corner which had a tiny bit of coke he tested in it, and he bought it lol the funniest part is the police intercepted the deal and he also went to prison lol.

I came here because I was looking to buy an industrial press and then it said: «people who buy this also buys: Benzocaine» and I was like «yeah these motherfuckers are making fake coke or they are cutting coke with benzocaine and pressing it somehow» 😂😂

I really love this channel. I'm a chemist as well, and it's fascinating to see syntheses and protocols that I have not had the occasion to do myself carried out on video . I would like to make one suggestion if I may. You don't have to cut down explanations to the absolute bare minimum. People that watch KZbin videos on organic synthesis are hungry for knowledge and they have a genuine desire to learn as much as possible. I think that your audience might appreciate it if you went into a little more detail on the hows and whys of the reactions. Not saying you do a bad job of it now and it certainly doesn't keep me from recommending your channel to other chemistry loving friends. It's just a thought.

Hey Michael would benzocaine powder 99% be a viable numbing agent because normal tattoo numbing creams aren't strong enough for me. And are there any health risk.

Great video, really helped me for my IB Chemistry class. Loved how clear all the explanations are and how you take into account safety precautions and concentrations to shift the equilibrium towards products. I will reproduce it for my internal assesment. Overall, excellent tutorial.

This is also used as a filler partly in cocaine so when they cut it with something inert it will still numb the gums giving the appearance of higher end coke.

Benzocaine don't cook the way coke does it's more like sand instead of a hard crack rock. So if the person that got the coke that was stepped on to make there own crack they'd find out fast and flip out on ya! Ppl get beat up or shot wen they step on it but if they only snort it they may not know the difference especially if they aren't used to doing coke often or it's there first time! Know your audience lmfao. Ijs

Thanks for posting this, I wouldn't have thought to look for benzocaine synthesis. I carried out the process outlined to create benzocaine for crystal growing. It worked very well!.

"Ignorant masses", I think that's a bit harsh. I actually started watching Nurd Rage when I knew very little about chemistry and it really got me more interested in the subject; even if it was on a very shallow level. At the time I would have been considered part of the masses--attracted to the flashy ka-booms in the videos. But now I've completed a bachelors degree in chemistry and am on my way to a PhD in chemistry. I definitely see what you're saying, but maybe there's value in impractical reactions if it will generate a greater interest in science (the world could always use more scientists!). Oh and Doug's Lab is good too.

I'd like to add 2 more: Tyler Dewitt - A great source for people learning about chemistry. (makes it really easy to understand) and Cody's lab - although I don't like watching him as much, he shows you can do a fair bit without a professional lab setup. (so maybe he doesn't belong in a 'Best' channel lineup..)

One thing to be aware of with molecular sieves is that they are slightly basic. This means that (in this type of reaction) if you add conc. sulfuric acid to them, they will be fairly likely to decompose to an amorphous sludge. They should still work (perhaps to a lesser degree, but i'm not certain) but it would involve a few additional filtration steps to remove the sieve powder. On a related note, don't do what I did and try to store your beautiful clean glacial acetic acid over 4A sieves.

I just did this with about a 76% yield. That's even after I had a foaming accident that lost several grams of product. I used a soxhlet extractor with molecular sieves to get the water out of the alcohol as it was refluxing. I used 40g (~0.292 mol) of PABA and about 250 ml ethanol, 15 cc of 98% sulfuric, and let it reflux for 2 hours. Then I added it to a saturated solution of sodium bicarbonate. I got too happy with the pipette, though, and that's when it foamed over. Washed it a couple times with distilled water, vacuum filtered it, and let it dry for 2 days in a sunny window. When it was nice and dry, it yielded 36.53g (~0.221 mol)

I did esterification my first year of chemistry but we didn't have 60-90 minutes for the reaction to take place. We also forgot to add the concentrated sulfuric acid as the catalyst. The reaction didn't go as planned but I learned a lot about esterification from watching your video, thank you!

Hot tip for higher yield... Reflux through a soxhlet extractor that is filled with 3A sieves, to remove water that shows up during the reaction. I'm averaging 86% of theoretical.

Also you can use a Dean Stark or a Soxhlet extractor packed with 4Ams (or 3Ams) to remove the water from the azeotrope and return anhyd solvent to push the equilibrium. However the rxn time will be increased as sieves aren't fast at absorbing H2O.

i had to just do a read up on words so here's the information for the random denizens of the interwebs i don't know the word cocaine comes from coca + ine (coca from cuca which was a group of people)...... so this information was lost i guess when cocaine becomes co-cane the substance was important due to the hunt for anaesthetics, before this point we could get them drunk or dope them on morphine the subsequent anaesthetics therefore inhert the "cane" part of the word, procane (branded novacane)..... they seem to bear some similarity (the ring and the double oxygen bond, the presence of nitrogen).... they have all therefore been categorised as amino-esters so in conclusion to this science, maybe you should try snorting some and seeing if you like it, it may have a similar effect to cocaine

As I'm watching this, I had to put a little on my tongue that I made about two years ago the exact same way. (hell of a toothache remedy) I used sodium hydroxide instead of sodium bicarbonate to bring the benzocaine out of solution. Thanks for the video and information. God Bless

How are Methyl Diethylamide or ethyl groups considered hydrophilic? If anything both ends of the caines shown would be hydrophobic and the center ester/amide would be the hydrophilic one. I imagine this would cause quite a weird confirmational change.

Interesting video. Now, is there a practical application? Would you mix it into maybe Vaseline to make a topical numbing agent? Maybe an ointment or cream for hemorrhoids? Maybe dissolve in some liquid to spray on for sunburn? In what ways would you NOT want to apply, (besides internally of course).

I once got benzocaine ear drops as a perscription from a walk in in clinic for an ear infection. When i went to get them, one place didn't have them, then they sent us to another pharmacy which only had 1 left and they said they hadn't seen it in a while. I used a little more than i should've by accident and felt very strange. I googled the drug and found it had been banned because of side effects in children. My head didn't feel right for the whole day, but thankfully i turned out ok.

Hey dude some suggestions on the neutralization of your acid to increase yield: 1) add the solid NaHCO3 thereby not adding any water to the rxn mix or 2) if you only have the aqueous base, add the acidic solution to the base while stirring. This way the acid is neutralized and hydrolysis of the product does not occur. If you add the bicarb to the acid slowly to not foam over, the pH is still well below 7 to conduct the acid catalyzed hydrolysis when you are adding water.

One quick question... at 1:11 the diagram shows benzocaine and lidocaine, but on the hydrophilic end of benzocaine there are no polar bonds, and there are polar bonds on the lipophilic end of the molecule, how does that work? Surely for something to be hydrophilic it needs polarity and equally so for lipophilic it has to be non-polar? Bit confused :S

Wouldn't slowly neutralizing hurt your yield? As you would be removing reactants from the equilibrium. Or would this not matter due to the neutralization of the sulfuric acid, it being the catalyst?

+Chase Wilbur Technically it shouldn't because like you said, the sulfuric acid would be first to be neutralized and it is the catalyst for the backwards reaction. I would say though, that I wouldn't be surprised if some was lost.

Would adding more sulfuric acid produce a better yield? Also, how do you know if you have your product at then end- is there a test you would recommend to identify esters or this molecule specificly?

The acid is just the catalyst. You could use less, but the reaction would take longer. Use more, and it won't speed things along very much. The yield would be roughly the same. To increase yield, you can pull out the water to keep the reaction from reversing. (but the reagents are relatively cheap, so why spend too much time and effort?) With reactions like this, the "end" is pretty much a judgment call.

Liked the presentation. 'Alone...' Funny. But feeling 'no pain,' or not being able to express same. For those that are interested, this might be a door into commercial, huh, chemistry? Purity is all. Heard about 2 Ph.D Chemists (brothers), who purchased technical grade dyes the spent a lot of time and effort to purify them. Then they resold them as analytical grade. They had more than a few bits of expensive gear to test with. And they kept good records too. So, then, good plan, good accountants, good gear, good reputation. Maybe good lawyers too? Maybe form acompany with the lawyers being business partners?

hey i’m new to this level of chemistry so help me out a little but are you referring to the C2H5 bonds on benzocaine as an ethyl radical? cause a true ethyl group would be C2H6 right? sorry i’m 5 years late hahah just confused and if ur still here i’d appreciate the clarity

You did actually address the reduction in concentration of the products, using 98% sulphuric acid means that any water formed is locked up by the reaction: H2SO4+H2O-->H3O(+) + HSO4(-) meaning it's unable to reaction with the ester like H2O does, that's the main catalytic property of the sulphuric acid as opposed to other strong acids. HCl works, but you get around 30% of the yield that you do with H2SO4.

This might be a dumb question but if benzocaine has an hydrophilic end why can you wash it by using water?(the same with the lidocaine one). Like i get that the salt form is hydrophilic but is'nt he clearly refering to the "pure" form? Thanks in advance

Could the H2SO4 be added via the reflux condenser? That way the solution would stay non-viscous, allowing the stir bar to stir. => No manual mixing required. And since we're already heating the solution, heat from H2SO4 addition would be less of an issue.

For anyone who cares: Toluene to nitrotoluene Separate p-nitrotoluene isomer Oxidize the methyl group of p-nitrotoluene using KMnO4 Reduce the nitro group of p-nitrobenzoic acid Fischer esterification of p-aminobenzoic acid to make ethyl p-aminobenzoate aka benzocaine Basically will have made a modern local anesthetic from carb cleaner lol
