how does affinity not change in non-competitive inhibitors? surely affinity decreases if the active site changes becasue of non-competitive inhibitors binding allosteric site.

I think somewhere in the lecture he said that substrates would still have affinity to bind with the enzymes, however it's so hard when Nancy the noncompetitive inhibitor is binding to allosteric site of enzyme..changing it's shape and making so many enzymes unable to bind substrates. However substrates still bind with whichever enzymes are available so the affinity hasn't changed which means [S] is also constant (we are assuming at half of Vmax)

My biggest question is how can uncompetitive inhibition affects Vmax and Km, km being the biggest brow raiser for me, yet Non-competative which can literally block a substrate from binding can only affect Vmax but not Km?????
