Really helpful to help me understand more deeply this technique. I'm from Vietnam so English is not my native language and studying biotechnology in the USA was really challenging for me. However, thanks for your videos, it makes a lot of sense for me to be clear to study this major and I graduated in Dec 2020. I watch your lecture videos everyday and still continue to watch them. My next step is to study graduate school to achieve my dream to become a scientist in biotech. I think it is because g-c bond is triple hydrogen bond making it more heat stable than a-t bond. And in pcr - high temperature exposure is present.... and that may be the reason high gc content used as primer .

One question , 72 degrees is the ideal temperature for the DNA polymerase so initially while increasing it to 94 degrees at 74 degrees do we observe change? like in the first step because the temperature will definitely be at 74 for a few seconds at least . Will the polymerase get acivated maybe?
