Thanks! So transfer to a water agar.... no nutrients for bacteria to feed on. But, the mycelium can still grow there and get stronger. Between your last video and this one, this is what I am hearing. I had not heard of water agar til you showed us how to make it. I think you are my fav creator right now. Helping me to understand agar work. Btway..... I poured 20 last night before I watched your vid. I didn't let it cool down enough so you know I have water on the surface. Anyway. Thanks for creating/teaching

Thank you so much for the support and for your generous tip-it truly means a lot! I’m thrilled to hear you’re enjoying the content and finding it helpful, especially with agar work. Your kind words really keep me motivated to keep creating and teaching! You’re absolutely right-mycelium is incredibly resilient. On water agar, it will often stretch out quickly in search of nutrients. This makes it an excellent tool for outrunning bacteria, as the mycelium usually gets a head start where you can snag a clean sample. I’m glad to hear you’re diving into these techniques-it’s a game-changer once you get the hang of it. As for pouring agar, we’ve all been there! Getting the timing right to avoid condensation can be tricky at first. The good news is, I’ll be filming a video soon covering proper pouring techniques and sharing some tips to combat surface water and condensation. Stay tuned-it’ll be up shortly! Thanks again for your amazing support and for sharing your journey-it’s awesome to hear about your progress. If you have any questions or want to share updates, I’d love to hear from you! 😊

@RealSimpleMushrooms that is fantastic! Both, the upcoming content and your being open for hearing updates and more feedback. You are welcome. When I find value in something that someone shares I try to grateful and mindful to award their efforts. You upped my agar game and inspired me to do my 1st pour 🫗 Well, actually I found you as I was pouring. Lol

@RealSimpleMushrooms and..... your thumbnails are rocking. The colored agar plates pop. And your vids look clean. Audio is good. Topic? Spot on with where i am in this agar game.

Happy New Year! THX for the video. I don't get the bigger picture. I'm just new to this. You said the Halo around your culture is an indicator for bacterial growth on the plate? That would be a nice detail I should keep in mind.

Not 100% of the time but it is an indicator

Is the trench just a cut in the agar or does there need to be a small gap? I find it hard to do anything other than a clean cut.
@RealSimpleMushrooms7 күн бұрын
You need to have the blade hot and turn it sideways to carve a gap with the broad side of the blade. It needs to be a clear gap all the way to the bare Petri dish.

Should I be using water agar in these processes or mea ? Many thanks I have been dealing with this for a while
@RealSimpleMushrooms7 күн бұрын
You can really use either. The meat will speed up the growth needed to achieve the hopefully clean transfer. But personally I prefer to make that transfer to water agar..then transfer one more time after that back to MEA

@@RealSimpleMushrooms thanks for the response! i put it back to water agar this morning , but i find the growth incredibly slow and very fine strands ... I will try both these methods on water next .... i was on T6 and it was making me crazy with the aseptic technique .....i was considering antibiotic agar , but i would rather avoid if possible
@RealSimpleMushrooms7 күн бұрын
You only need one of those fine hairs ;)
@RealSimpleMushrooms7 күн бұрын
When I put something to water agar usually it will go rhizo (but be very faint) and as soon as it shoots off from the transfer I’ll grab a piece and toss it back onto MEA
