Do I have to have graphing tape to seal up my plates once I've transferred, or can I use the tape Ive got on hand which is micropore. Thanks for all your help. I'm loving this new hobby and you have taught me most of it.

Micropore will work if that’s all you have but the plates will dry out pretty quickly. I wouldn’t recommend it for long term storage

NGL I almost skipped through this video because I have been watching a LOT of videos on agar and I didn't because I heard experience in your low-no BS-tone of a voice, plus you have a really clean and inviting environment, plus your appearance says a lot more about the information you provide. This is thee most informative video I've come across and I've been "learning" from Mossy Creek, North Spore, Spore n Sprout among many many more and yet I've learned more half way through this video than all the other hours spent elsewhere. We're a nonprofit, just starting out and it's been a one person show for 6 months, I have no major clue what I'm doing so I must rely on videos like these for things I can't find in books and I'm about to place an order to get all of these supplies. I have what appears to be all the right things in my shopping cart according to all of the items you have listed and that makes me happy to know I'm in the right place to get started. Thank you.

If you had a multi spore syringe and wanted to put the spores on agar to see if they are clean What would be your technique for doing that? And I'm guessing the potato agar plate would be the plate you use. Or is it the water agar. Thank You for your Help.

It probably leaking out the way because of all the pressure it has to release pressure somewhere what does it have a way of releasing pressure out of the machine? Also I'm not sure but I think that the wallpaper steamers have a extra little hose inside the hose that backs off pressure when you press it against the wall too hard so the pressure don't build up in the machine because if not It could probably mess it up. I'm not sure but I thought I noticed that on another video.(Or does this steamer also?) I'm looking forward to making one so any insight would be excellent. Thanks 🙏🏻

Do you think it's worth buying a ink bird controller to turn it off when it gets up to temp than back on when it drops so you don't have to watch it? You can even get a wifi one that will notify your cell phone. I actually have one but it's not WiFi 😢

For that to be 100% accurate you'd have to bury the sensor in the core of one of the bags, it's certainly crossed my mind. I wish they had like a spike probe for those things.

Yep, just move the lids with one hand and pour with the other, whatever feels natural to you and whichever way you can move the smoothest

So, because I have a flow hood I actually don’t use the sleeve and keep my plates in front of the hood until they are cool and all the condensation has gone away. I don’t usually use the boiling water trick anymore because I have a hood. When I was working in an SAB once I was done pouring my plates I would put the sleeves back on them and then remove the SAB and put the mason jars of hot water on them and leave them until they cool completely

I was also curious about this. My flow hood is inside a sealed lab that is inside another lab that has an airlock to get into. I'm always worried about leaving my flow hood running for some reason but I also never put the sleeves back on until after they cool. I can't wait to try this new method. Thanks for the vid.

Thank you for the video! You mention "leaving the plates until the next day". Is this in front of a running flowhood overnight or is it turned off after pouring? I wonder if contaminated air could get sucked in while the temperature drops. Do your petri dishes have ventilation nocks?

I leave my hood running, if not I would just cover them in the sleeve and keep them in there. They'll be fine.

Thanks for the vid!! Question, my LCs I’ve made which have Nutritional Yeast are instantly cloudy before even inoculating and after sterilizing. How would you tell contamination then? Recent LCs I’ve taken the NY out and no problems now however, I see random “floaties” on top, once I stir, it disappears

that's normal, once the mycelium starts to feed on the nutrients in the jar and grow you should notice that cloudiness subside, the jars here with both inoculated the same day.

@RealSimpleMushrooms I've been using grafting tape for a while now, since it has pretty similar properties to parafilm, at least the ones we need. (I come from a fruit growing background). Your videos are very informative, devoid of speculation and completely in line with the theory, maximally simplified for a wider audience, and at the same time entertaining, which makes the information easy to understand and adopt. I grow a bunch of oyster and get some crazy looking ones depending on my CO2 and humidity as well as whether they grow from the tops or sides of the containers. I do pasteurized straw in just about any bag or plastic bottle I can find. I have cloned and swabbed a bunch of them but so far they don't seem to be traits that carry on.

@@RealSimpleMushrooms What do you grow your lions mane on. I do hardwood sawdust from my mill (I have tried all different mixes) enhanced with wheat bran and gypsum. I get constant harvests of dense fruits about the size of pool balls but would love to see some giants.

Mine ended up being all over the place to finally give me the correct humidity without running all day. Have you seen pics of the King Oysters grown in bottles? Some of them get pretty horse wankery.

All part of a plan, I didn’t post for a very long time and I’ve been working on the SEO on the channel so I’m posting 10 videos to give the algorithm a bunch of audience data so it can recommend my videos more and to the right people. It’s been working so far. Every video does better the the prior one. Once I hit that 10 video mark I’m going to get on a schedule of either weekly or bi-weekly with maybe some live stuff sprinkled in

If you have a gas exchange hole or are using a lab media bottle you can screw them in tight, if you just use the metal lid upside down you need to crack that one loose a bit. And yes I have them slightly submerged…maybe an inch or so. Glad you’re enjoying the content. My follow up to this video is scheduled to go live at 3pm est today

What makes a "true casing layer" if what you did on top isnt??

A “true” casing layer is a layer of 50/50 peat moss/vermiculite. Peat can hold 20x its weight in water and the mixture is non nutritious. It helps with humidity and moisture retention

Thanks! So transfer to a water agar.... no nutrients for bacteria to feed on. But, the mycelium can still grow there and get stronger. Between your last video and this one, this is what I am hearing. I had not heard of water agar til you showed us how to make it. I think you are my fav creator right now. Helping me to understand agar work. Btway..... I poured 20 last night before I watched your vid. I didn't let it cool down enough so you know I have water on the surface. Anyway. Thanks for creating/teaching

The water agar is a good idea. Btw, if you have an Asian market near you, agar is pretty cheap. You can also get packets for single use... if you're doing a good amount of agar work lol. They're like 25g or so

Is the trench just a cut in the agar or does there need to be a small gap? I find it hard to do anything other than a clean cut.

You need to have the blade hot and turn it sideways to carve a gap with the broad side of the blade. It needs to be a clear gap all the way to the bare Petri dish.

You can really use either. The meat will speed up the growth needed to achieve the hopefully clean transfer. But personally I prefer to make that transfer to water agar..then transfer one more time after that back to MEA

I will try both these methods on water next .... i was on T6 and it was making me crazy with the aseptic technique .....i was considering antibiotic agar , but i would rather avoid if possible

When I put something to water agar usually it will go rhizo (but be very faint) and as soon as it shoots off from the transfer I’ll grab a piece and toss it back onto MEA
