I don't understand though ... I understand most of it, but why does the salt start to react with water at higher concentrations. It's not as if the ionic interaction between the salt and the negatively charged residues isn't still stronger than the bond between the negative side of the polar water molecules and the positive salt. Intuitively, I'd think that the salt would first interact with the protein, and then when there isn't enough left, it would then react with the water. What am I missing. Can someone explain this to me? Other than that, I understand this.

sir can you please tell me why and how high molecular weight has less surface area and that leads to less hydration less salt required for precipitaion of protein in case of globulin which is know as half saturation and which is exact opposite for albumin that ppt in full saturation

Beautifully explained thank you! I've been struggling with sample preparation for months now - I have VERY expensive triple-labelled samples for NMR so, naturally, sample stability has been a great concern to ensure we can get the most out of each prep. Have you got any rough kind of buffer optimisation "workflow" that you go through when determining protein solubility? I've thus far tried crude hanging drop precipitate screens using a crystallographers microscope, as well as a thermal denaturation assay utilising a hydrophobic residue-binding dye which infers protein unfolding using qPCR (ThermoFluor I believe is the technique name). Have you got any other suggestions? I'm still struggling somewhat with long term stability.

I just wanted to know if it's safe to salt steak on the grill. 🙂 Actually, I suspect I'm Fragile X positive, but I can't get the test. Insurance is ok with it. I just can't find a lab. So I'm thinking, "I have a microscope...and lancet...and leishman's dye...and salt." So I figured I could give myself the test, but it appears more complicated than I expected. I do that. I oversimplify things and then realize I should've gone to a good college in persuit of a PhD (dang dyscalcula!)...but I probably would've got lost on the way to college, anyway...(dang dysgeographia!) Oh! ... Nice presentation, though! 🙂❤️

Great video! Just have a litte remark: At 1:52 you mark the carboxylate ion which is part of the protein peptide backbone. Instead you should have marked the carboxylate at the amino acid side chain which is exposed to the enviroment.

Ammonium sulphate precipitation follows the procedure explained by you. It is the good and simple method instead of ultracentrifugation and other methods. Nicely explained. Continue.

Great bro. Could you talk about why Lanthanum.Cl3.H20 (anhydrase) precipitate in room temperature water? (it has low solubility for sure). This question has nothing to do with protein solubility.

I think with high molucal weight..we in are in case water in molecule. When the molecule is very low weigheit we are in case in molecule in water.. I think in hight salt the high molecule began to contract and push water in outside. We can see that in milk coagulation by elevating temperature.. I think all high molecular product can forme an emulsion Water in oil or oil in water.. Thank you for video..👍👍🇩🇿🇩🇿

From your video, I understand that adding salt ions can make the structure of the protein stable along with reducing of the intermolecular replusion of the protein. Then, what is the mechanism of the water molecular can form the hydrate shell around by the protein? Is hydrogen bonding on the hydrophilic regions of the protein? Thank you!!!!

however i have a question, regarding the protein isolation, if i not sure about the solubility strength of my desire protein, then salting in (addding low concentration of salt solution to allow it to solube is better) or salting out is better. And base on explanation, i understand that both salting in and out is helping to isolate, is there any condition like protein that not likely to soluble will be favor for salting out process? if there is, is the example fibrous protein will be more easier to seperate in salting out or salting process? I understand salting in and salting out based on their definitions, but I don't understand why at 1:33 the definitions are placed on either side of the graph like they are? I would assume salting in would be in the left side, and salting out on the right? Edit: There is a clearer graph at 4:53 with the definitions on the "correct" side.

This video neglects to show the chloride ion in salt, NaCl. The logic of the video no longer makes sense when the Cl- ions are imagined in the diagrams.
