I use oven tek for my agar pours. Preheat to 250, crack it open and place a wire rack over the opening. Do your pours over the rack, good to go. Learned it from PGT. Thanks for the video Willy, good info for sure

I do mycology in the closet. No air current. I have pretty good results. With safe food handling practices. Bleach water spray bottle is my go to, sanitation for pennies, millet bird seed it the grain of choice you can get like a 30# bag for like $14, you can find damaged bags for like $3 at lowes and the depot stores.

I use my 2nd wardrobe closet no clothes, storage products, no vents= no airflow- possible contam… I use the top shelf and got a few of the Boomer SHROOMER inflatable monotub. It took me a few weeks to find my groove getting my grains to not turn into contam. I barely go into my closet lab space, unless I’m working, and I take a shower and turn ac off and spray my gloves, forearms, and upper torso with 70% iso. Also I use a plastic tote that melted 8 holes the diameter of a Campbell soups can heated up red hot, and carefully apply pressure into the tote for the holes to be melted in perfect circles no drill debris of plastic shavings. I’m still learning but I finally got it down now. Next step I’m making my own DIY Mini laminar flow hood all for under $80

Thanks! I like this, what I have done is take a similar sized bin, and wrapped shipping saran wrap around the top 2/3, leaving a wide hand port. I make it clean, but my bunsen into it for 10" and go. So far, so good.

Hah! I just saw this and I’ve been using my box exactly like this but I added an extra little safeguard. I added a cut piece of clear plastic tarp to cover the majority of the front part with a slit cut 1/2 way up so my arms can go inside freely.

I've had great success(10 agar transfers and 4 grain transfers with no contamination) by using this sab method and I am happy you posted about it because everyone thinks im crazy for not cutting arm holes. I started doing it a few months ago because I realized during a smoke test that when I put my hand into a sab(through holes) air is sucked in and it does the same thing when you exit. The trick is you have to have the opening flush with the edge of your kitchen counter so you arm movements do not push air into the counter and into the sab. You also cannot have any airflow pointed towards the opening. I also sanitize my hands and arms every time I leave the sab even if its for a second. If I am working more than 15 min then I spray lysol into the bin. You can spray even if there is stuff in there as long as none of your plates or jars are open. I actually think this is more effective than the traditional SAB with arm holes. The armholes are great if you do not leave the sab until you are done but if you have to exit for any reason then when you re-enter you push air from outside into the sab(from air displacement) directly over your samples. If you are disciplined with your movements (always reach from below or level with your samples) and with sanitizing using the open SAB is more effective.

Hello. How to sterilize a scalpel and an inoculation loop with an open fire in such a box? will fire create air movement or not? besides, it's flammable, because lysol contains alcohol, isn't it? Please help, I can't figure out this issue with the sterilization of a metal instruments.

@@СквиртомирЯросветов I use a lab grade alcohol lamp too sterilize my tools. I do not keep the lamp in the SAB. I leave a few feet of open counter next to the SAB and sterilize it with alcohol and lysol before placing my lamp right next to the sab. Sterilize your instruments then while they are still red hot transition to working in the sab. Smooth but urgent movements are key to prevent contamination. Plan and practice everything you are going to do in the sab before you do it. This technique isn't perfect and there is still a risk of contam no matter how good you are. I have since upgraded my setup to a labrats laminar flow hood. Edit: let the lysol and alcohol settle/evaporate before you light anything outside the sab. There can be fumes inside the while you work but I always extinguish candle before I spray.

I take a 10 gal aquarium, turn it on its side. Then I tape a towel to the top edge and two sides leaving the bottom with the slack of the towel.

For your SAB holes all you have to do is put duct tape around the holes or glue a plastic tube around the holes to help your arms

instead of cutting the holes find a metal can about the size of your arms, heat it up on the stove and burn the holes through. leaves a much smoother finish and a perfect circle with less chance of cracking the tub.

Hello, just starting out and wondering if this method still works for you and is still your go to? I tried it and got decent results but I am new and those results could be from any contam point. I switched to a SAB and seems to be about 1-2 out of 10 grain jars contam. But they did not contam on matched AGAR samples I kept. I intend to go back to Your Idea but try all over now that I have more knowledge than before. Thats why I was asking what you still think a year later. Is it worth it to use the homemade flow hood? Obvious there is way better but I mean on par with your statement.

Another idea to add is Saran Wrap across the front. Leave a opening at the bottom. Easy to get your arms in to work and easy to see through. Keeps the work space a little cleaner

@happygolucky1100 I’ve used this cheap very effective method without l the hassle of buying very expensive equipment that you don’t necessarily need but they are very good to have to eliminate altogether or reduce contamination

I saw a trick where someone put their oven on very low and had a wire trey reaching out over a slightly open oven. The theory was that the rising heat created a somewhat laminar flow upwards and he poured his agar that way with the dishes on the wire trey. I have a sab so I don't need to do that. But it sounds like a good idea for broke boy tech. Haha. It would be cool to see some comparative results with these different techniques to see if one has less contam rates then the other.

I've done that oven tech 4 times now making plates and slants and it works surprisingly well. I usually pour 4 cups of agar each time. I've been using the 2 ounce plastic condiment cups so I come out with quite a few. Probably about 50 but I usually do a fair amount of slants too which seem to have come out clean. Last pour I only have had one plastic cup with a little contamination so far. 25 cups I did of a separate syringe all came out dark brown everywhere the loop sneaked across the agar and it was fast. Second day I was throwing them away. The scary part was that I used the same loop for 4 different species one right after another. So we know that flame sterilizing does a excellent job.

I've only done it once right in front of the hot oven, advised by midwest. I got the kit from them. Have you heard of of oven method? After seeing this it does seem better than the oven method. Yup I use a c-clap to keep rack secure. I learnt from Home Mycology. Taping a clear dry cleaning bag to small hepa fan works really nice as well. Makes huge sterile area to work

Working over a cracked oven with a rack also works nicely, shout out to Home Mycology. Have also taped open end of clear dry cleaning bag to HEPA fan. Turn on and cut arm holes in side, balloons out creating huge sterile area to work

"Cuts arms" omg that made me laugh. People with that little resolve might not make it in this hobby. The only reason I don't like a SAB is because I'm disabled from labor jobs due to my back. No worries I got a work from home job, as not working is the worst thing for anyone. The sab is horrible for posture and long sessions are not a viable option for me long term.

Lowes sells a small plastic greenhouse for 30 bucks. You put the frame together and slide the plastic shell over the top and cut 2 arm holes in it. Works great

Thank you for your videos! It’s very interesting, you doing amazing job! Can you do video how to get 2nd and 3d flash for small commercial farm? I just started and can’t find how to do in KZbin. Not sure if I have to soak each bag for 12 hours? It’s a lot of work, is there is a way to do without soak in them and get 2-3 flashes?

I never made a SAB. I took a tub and just turned it on its side and sprayed isopropyl alcohol on absolutely everything I touched and transferred to/from. Keep open flames away from that environment if you follow my lead. I've only had contam in a few agar cups. Transfers from agar to jars or jars to tubs have not had any contam. Not sure why only a few agar cups have contam and nothing else has. I suspect maybe syringes were contaminated?

Funny how no one in the community seems to appreciate the open flame technique, which is btw the standard in academic microbiology. Typicaly it would require a bunsen burner and a source of gas, but I work with fondue fuel and the lil burner that comes along and i get pretty decent results, working in the sterility cone. I'll admit I'm using PastyWhite's no pour agar technique though.

I was using an inoculation loop to start 4 different species on agar. After about 3 days I checked them and one species I had about 25 agar cups of was growing dark brown everywhere I streaked the agar plate with the inoculation loop. No white mycelium, just dark brown. The other 3 species have come out really clean and healthy. The ones that came out brown and contaminated where the first ones I did. Did all this in the same SAB Box, same batch of agar, with the same inoculation loop. If I didn't flame sterilize the scalpel, that would happened to everything. I only flame sterilized between each species. I really think the syringe is contaminated but I made more agar cups and put the spore solution on the agar directly from the syringe just to reduce as much contact as possible just to see if it happens again. Also flame sterilized the needle so I can hopefully narrow down what caused such an aggressive contamination. It took over 80 percent of the agar surface in just 2 days. It will be interesting to see if it happens the second time.

Also I gorilla taped and cut a small cross with a razor blade into two sandwich bags over the arm holes which were just cut bigger so i didn't cut myself. This keeps my feom cutting myself and keeps air from getting in when i put my arms in and out... And i have done the same thing, but with clear industrial strength Saran wrap over the top half but tbh the alcohol lamp just worked better for me as long as you work close. You just need heat to push it all away. Also you can build a laminar for about 80-100 bucks its just the right fan behind a HEPA and enclosed, im even going to add some UV lights

I use to grow mushrooms using the hydrogen peroxide method. Helps cut down on contam and can work in open air.

You can make a flow hood for 40-100$. Only way to go. Either box fan with home filter taped to it. Or an eBay mounto hepa filter with an old squirrel cage and build a box. Hot glue works to seal shotty wood work 👍🏾go from 5-10% contam to 0. I put it in an ice shanty to recirculate clean air

im still figuring stuff out, the kit i have had me innoculate jars in front of my oven at full blast with the door down and doing it right on the door it self or holding in front of it at least. does this not work for things like agar and syringe and prints i take it? just curious, as it would be the most convenient i feel.

What's your thoughts on using charcoal filtration system instead flow hood? Unfortunately it is setup to recirculate. I'm going to exhaust it out window so it'll be negative air pressure? I'm novice/ beginner. Thank you for teaching me new things the correct way. I found new agar plates & ready bags with contamination 💩💔 seems like agar containers should snap r screw on 🤘😎🍄

Carbon won't work for this. It does chemical filtration, removes molecular sized contaminants from the air. You need mechanical filtration, like a HEPA filter, to remove the larger bacteria/fungus spores from the air.

This is exactly what I did a week ago on a spawn to spawn transfer! Except I was also running 2 UVC w/ ozone lights inside the box. I’ve killed contamination with these lights. A must have!!

Please be careful with uv. My homie got wicked face/neck sunburn and couldn’t open his eyes for a day. He didn’t know it was on under a table, was out of sight. Had a remote and usually turned off hour before entering. He was in the room just over half hour and messed him up bad. Just be safe with them

I did some petri dishes. All are gorgeous. One had a orange spot growing. I put methylene blue on the spot. It seems to killed the orange. Jack frost is doing great 👍

I got some spores and put them to agar. The problem is they were delivered early and sat in the mailbox for 2 days. It is super hot here and I'm concerned that by not knowing they were here, I unknowingly screwed up and they probably aren't viable. Golden Teacher and Koh Samui are the strains. I am just learning from you and mycotrophic and I hate to think I wasted my $. Can you tell me your thoughts?

you probably wont get a 100% success rate but its better than nothing and you can get it pretty high if you are careful. a laminar flow hood is just too expensive for a lot of people

You do an initial wipe down with a sanitizer, then coat the inside with a fine mist of water (ideally with very little soap) to trap particulates floating around in the box

I spent 15 bucks on temu and got this massive collapsible still air box where the sides zip and unzip and there are armholes with sleeves and elastic wrist cuffs and i do this exact method. I Lysol it, let it sit, put my shit inside and spray with lysol one mo' again and once its settled ill untie the sleeves, spray my hands andfet to work

This is 2mo late but I believe it’s because those containers can’t go into the pressure cooker. If they were PP5, you should reuse them.

Just a tip for people that live in a newly constructed home: your house, most likely, doesn’t have enough contaminants built up to affect your grows. So you don’t even need a SAB or flow hood. Just do your sterile work in open air. But make sure you change your home air filters regularly and to turn off the HVAC system about an hour prior to working. Also, have good sanitation practices!

Unfortunately, this is bad advice and even worse procedure. The air, wherever your room is, contains microbes of all kinds which cause contamination. Lysol is a disinfectant, not a steriliser, does not result in asceptic workspace, especially open to air movement like your open plastic box.

This ain't rocket science. Make it make sense. Dollar store pool noodle cut to circumference of the arm holes, then cut long ways to the center of the noodles. Place around the arm hole edges. Skin cutting Problem solved...

FYI shroomery has long advised against using lysol because it is petroleum based and is believed to potentially cause mutations. Lots of people still use it, but I think bleach or 70% ISO is a better option. Remember, there's zero chance of actually creating a sterile environment like this, you're just trying to reduce the chances of potential contamination.
