Empty vector is linear, which means that it cannot replicate inside E.coli. Normally vector cannot ligate onto itself because it is cut with different enzymes, so it has mismatching sticky ends. Sometimes vector digestion is incomplete, so it does ligate onto itself and replicates. This causes high background levels. That's why we have control plate when we do transformation.

Can you again please explain why we do not have colonies on the control plate, as you didn't use any antibiotics. Or the competence cells that you use do not grow without presence of the vector? Thank you.

Just a little confused here. The vector has an antibiotic resistance gene right? So it empty vector is taken up it will still grow?
