Hi Ed, on this you are using the same mono to cross as an example; I think? So if I follow correctly, If I have your AG or RW+TOQ isolated as a mono without clamps; and I plate it with the MK+TATBC (assuming I successfully isolated that mon)....that I could possibly mate those two? I think I get it; but the 2 LSGs on screen throwing me a bit...Thanks

Almost any two monokaryons will be compatible with each other. There is a small possibility that two monos from the same fruit (LSG) will be incompatible, but you don't need to worry about it (mating alleles and some other B.S.). The reason I mated the two LSG monos was to prove that they weren't isolated contaminants. If you don't have experience on the scope, it is easy to confuse a contaminant (which won't have clamps) with a mushroom monokaryon. Monos will not make fruit. So, if you take two monos, let them mate on a plate, and see fruit, you can be assured they are both mushroom monos. If they don't make fruit at the interface, it could be for several other reasons.
