I'm trying the bucket method on Blue Oysters and I did it wrong on pine with lol 7 buckets. Live & Learn. So trying again with different buckets and Aspen wood. Wondering if dumping 5 galleons of boiling water on the Aspen shavings and letting them cool or letting them cool till cool enough to handle is the right thing to do. Kinda murky on the difference between pasteurization and Sterilization here. I know milk get flash heated to almost 300F then quickly cooled down and that denatures the proteins and I'm thinking that is Pasteurization. But Sterilization is long term hard core High (boiling hot) heat to kill everything. Right? This is my first foray into growing via the bucket method. I just bought some of your brown treasure and then going to try that with a popular dung loving species. Getting my feet wet here. Grateful for all you do. So passionate, fun, generous, easy to understand and downright awesome and KIND.

Thank you for the video, is it possible to put agar cut into liquid culture medium to let the mycelium grow to make lcs?

I was gonna make my own flow hood. I was gonna put a ultraviolet light inside the box and turn it for about ten or fifteen minutes before work. Does this sound like it would help keep contamination down? Looking for an opinion on if this is a good idea or a waste of time and money. I haven't seen this done yet?

Do not swipe your agar plate all over the place like Willy, keep one small inoculation at the center of the plate (you want the culture growing outward from a single point, and you will take cuttings from the outer edge of growth before it hits the side of the plate). And its really not very hard to use a sterile (flame sterilized) xacto knife or scalpel to cut a small piece of agar out, practice on a spare agar plate, takes 5 minutes to learn to do it easily. Using a hole punch seems a bit ridiculous to me and its going to be harder to keep that clean (vs. a scalpel) and it probably won't be as sharp.

Hey Willy, awesome Edu here. Appreciate all your enthusiasm, tips, tricks, passion and love! So ya a question? Can you explain the difference between Pasteurization and Sterilization? inspirations
@jessicahay9305 Жыл бұрын
Sterilization kills everything living in/on something. No bacteria, mold, microbes etc. Pasteurization kills most of it but not all.

Yea he needs to smear .and that loop isn't used like that it has a cutting edge to scoop out the dish it makes a little scoop thrn you can just transfer it but I'd definatly smear soci could see growth and have wider parameters to transfer he over top him self and talking lc should be done with a syringe it's it's off a slant I'd definatly use a swab ...and a little multiplayer like a soy peptide ...squiggle is right he knows his stuff but you could use the centerdudge bendil and Swerell like a lc and thrn swab off that bit remember sterile teck...

Love your content bro, I think the first videos I watched before ever starting. But, I gotta say, I was freaking out inside every time you set your dish lids in front of your dish. Should always open and move away from your airflow, don't work between the filter and your open plates.
