yea it kind of breaks my heart. the reason why is because claire made a video talking about getting her lung transplant, i specifically remember her mentioning her future thinking she never would have one and the sense of anxiety she was starting to have thinking about it. it was scary knowing she could live when she was told she was going to die young. and that small glimpse of feeling she had is what breaks me when i think about her.

Yes!! This is why we have so much hope. My son is 4 and will be able to start this in the next couple years, once the FDA approves it for 6 and up. He is currently on Orkambi and doing SO well, but Trikafta is what we have ALL been waiting for. I feel like I'm living in a dream, we are so lucky to live in this day and age 💜💜💜

@@mcmelre that's so incredible to hear, to know that your child will have a true fighting chance & not have all the awful organ damage. I deal w smth dif,.. I had lupus for years but since there's not a treatment that 'treats' v well (there is one but it's $70k/yr), things have progressed (even while on a dif med that slows progression yay ugh) & I now have around 5-6 autoimmune diseases + symptoms from others. ...MY POINT - my daughter is 15 & I'm having her tested for AI issues bc she has problems similar to me that no one else she knows has. She's much taller than me and grew quickly so for years we wrote her pain off as legit growing pains but now I'm not sure. If only we AI patients had a medicine to take that really did anything, that would be so reassuring. Esp since my new dx is dif than lupus. It's rare, serious, shortens life expectancy, & a basically guaranteed (barring an accident or smth random) that I will die from it. And there's no telling when:/ Everyone deserves a full & healthy life w happiness & I'm so truly thrilled for you & your child, for your family & friends, and everybody who gets to take this new drug! 💜

bare bohemian I understand you completely. I’m an autoimmuney too, I have AS, crohns, gastroparesis, chronic pancreatitis and exocrine insufficiency... where’s there’s 1 theres 5, right... a fun little group then with a whole host of little things not listed, you know how it goes, unfortunately. I wish there was a magic pill for autoimmunity. I’m on remicade methotrexate sulfasalazine AND prednisone and still get hospitalized for at least a week at least every other month. All they do is symptom treatment, I wish with all my heart they’ll make some headway and figure things out for us. I’m so happy they have found hope for CFers. I wish your daughter a good outcome and you the very best. Stay strong and don’t give up, I will keep you in my thoughts. Sending much love.

I have been on Trikafta since October 2018. My story sounds very similar to yours, I had been in survival mode for years just trying to keep afloat. I had started transplant workup when I was put on the initial double blind trial. Within hours of taking the first dose I couldn't stop coughing up, it was like I just couldn't keep the mucus in. After 36 hours I stopped coughing all together. Fast forward a year I have started working full time (I am 30 and have never worked full time in my life) I got married earlier this year and didn't cough once on the day. My wedding video has no coughing and I danced for hours . The improvement in my quality of life is amazing I can't put it into words. I can't believe how lucky I am to have been given this miracle drug. I never thought I would have these results. I'm so happy that you were able to get access to this drug and I hope you continue to have amazing improvements.

Thank you for making this video. I'm a scientist in the biopharma industry. In moments of frustration, we try to remind ourselves that our work has purpose and makes a real difference in patient's lives, but when you never see the results, it's easy to become cynical and jaded. For every one drug that makes it through clinical trial and gets FDA approval, there are hundreds that fail somewhere along the way. To spend years working on something only to have it be "for nothing" can be soul crushing. It's nice to be reminded of the successes.

Thank you for what you do. It's such a blessing to have hope and a longer potential future because of all that trial and error, time spent and so much more. I'm not a CF patient, but I suffer from a rare cystic lung disease and now have a treatment that wasn't the case 15 years ago. Not as drastic of improvement in function, but simply less decline. It's nothing short of a miracle.

I’m both so incredibly happy for all the CF patients I see finally getting a breakthrough. Can’t help but think about Claire Wineland, I wish so much she was around to be able to try it. But she raised so much awareness and touched so many people, I can’t help but believe she was a catalyst for this.

V520X would be a nonsense mutation - "X" means the letter V was replaced with a stop signal, which we call a nonsense mutation :)

This may sound weird. We had a really bad year medically: we being my disabled son as patient a d me as mother/ caregiver. And with his illnesses there's no stuff on the horizon that's even close to clinical studies. It's all empirical: if it kinda works, fine, if not try something else ... Yet I find this video so extremely encouraging. It makes me so, so hopeful to see such improvement with someone else and with a totally unrelated illness. But it reminds me how grateful I am to live today and what changes may still come. Thank you so much for letting us witness this special journey of yours! All the best wishes from Susanne

Yesterday's vlog Mary helped Peter move that gigantic bookcase upstairs. The lung function and accompanying stamina is mind blowing. A year ago it wouldn't have been possible. I think it's true as you'd said Mary seems almost at the healthier point before Scottland. I look forward to this new season of healthier days. What new adventures await.

I've been watching for about 3-4 years now and almost every single vlog always used to contain a coughing fit. I always felt so bad seeing you like that, you are such a freaking trooper. Peter being your caregiver, he's just simply amazing in being your rock. I love that your quality of life is so much better and your not in survival mode anymore! So happy for you guys! Hi you guys! Greetings from Germany! I'm a 31 year old mother of two boys with CF (6 and nearly 2 years old - both diagnosed within the first two months of life). Since I have heard of Trikafta and the amazing study results and of course of the successful approval in the US I am absolutely keen on reading and finding any kind of news about this drug. The latest developments feels for us parents a little bit as an early x-mas gift and so we have lots of hopes for the next maybe one or two years (whether it would be approved here in Europe or especially Germany, too). This is something I dreamed of for my kids from the very first beginning. Thanx a lot for your reports and I wish you all but the best for your further future!!

Ive been extremely busy so haven’t seen a single upload from this channel in months and I AM SHOCKED BY HER IMPROVEMENT. Literally shocked. This was on my recommended and after watching a few seconds I had to double check that this was the same person I had watched a year ago. Her voice, energy, color, everything about her is different. She looks like a completely different person honestly. For example, at 5:50, I’ve never heard her spontaneously uproariously laugh like that with especially with no coughing afterwards. Her speech and mannerisms were always very measured and muted. That laugh was amazing. So happy for you!!!

Completely off topic but what temperature was your house when you filmed this? Mary has a thin short sleeve shirt on and Peter has an undershirt, long sleeve button up shirt and a coat on top😂 No but seriously....so glad to see how well you are doing. We all saw an immediate change in your energy, coughing and less raspy voice due to less mucus and coughing. So happy that this drug is now available to other CF patients now too💕

As a retired special education teacher, I was in college in the years 1978-1983. Children with CF were expected to live age 10- 12 years old. It’s AMAZING how research has helped you and the CF world! I’m thankful that the Lord has kept you Mary healthy enough and able to benefit! You look and sound healthier!

The other day a video was suggested for me to watch. It was about a CF patient that has liver disease and doesn't think she can take the triple combo. I am so happy for you Mary! I started a new medication called Fasenra for severe asthma. My lungs are clearer. It’s a wonderful medication for me. My quality of life is better. I am so grateful for my respiratory doctor fighting my insurance for this medication. I have been on it for 9 months. So far so good!

I’m so happy Trikafta is working so well for you! I should be getting on it within the next few weeks and I’m so freaking excited!!! My mutations are DeltaF508 and G551D. I’ve been on Kalydeco since 2012, and I truly believe that drug has kept me from needing a lung transplant. I used to have to go in the hospital 1-2 times a year, and now I haven’t had to go into the hospital for a CF exacerbation in almost 5 years! My lungs haven’t been feeling very well for several months now, so I think I’m gonna need a tune-up very soon, but I’m okay with that.

Now i see on sci-show Hank speaking about this wonder drug and i instantly remember about you. I dropped my fingers on keyboard and typed my way back here ... I have epilepsy and the I went through an adjustment when I found the right medication. Once my body got used to the medication, the side effects calmed down and things got better over time and I gained so much quality of life. I still manage the side effects, but it's like Peter said, the quality of life gained is SO worth it for the side effects, at least in my case. Very happy for you Mary, you guys are the best

I too have epilepsy. I somewhat denied it for a very long time, thinking it was just a quirk that happened, until I didn't take the medication for 3 days. I was found in a post-lictal state, the period that follows after a seizure, by my daughter 5 months ago. I never wanted to give into this diagnosis or give it a name because I didn't want a lifetime of medicine and sigma that comes with it. Now with this new hope for CF'ers there seems like there is a real hope with epilepsy too. 🙏🙏♥️

im on day 5 and i feel amazing. i felt so much better after day 2. with symdeko it took me about 7-8 months to really start seeing results. this med is such a game changer!!

I wonder...if ur body will produce less & less mucous over time (ie cumulatively less)? If that happens, I also wonder if those antibiotic resistant bugs will then have no place to live in ur lungs (& be eradicated), &/or ur immune system may have a chance at eradicating those bugs? I know potentially a bit of a pipe dream, a “wacky” theory, but my mind has been going there, with u having such an incredible response!!! It’s beautiful to see how much more u “Trikafta can” these days! Just INCREDIBLE!! I’m SO unbelievably happy for u both!! I know u & others are SO thankful for Trikafta+their brilliant, wonderful & incredible responses to it-but I hope & pray this disease modifier is “just the beginning”-that a CURE is on the horizon & not long away!!!
