Unbelievable! I was working in a hood subbing orchids in 1983. Then to big scaled up industrial size propagation for export, then labs, and genetic modifcation shooting genes into callus with a modified gun. Then virus elimination for quarantine germplasm. Yet after all this I click on this little old vid and I still learn something new. Never seen a roasting bag used for sterile transfer. Never knew it was sterile. thanks for this upload. It's a pearl.

I really enjoyed this video. I'm starting to experiment with tissue culture but I know it's pretty tough to get it going the right way, using different growing media's and nutrient hormones etc.. This is so interesting and I just love the science behind all different walks of nature. Thank you for sharing! I wish I could visit a lab that does this. So fascinating.

Damn it. I just spent $500 on a small laminar flow hood and I could have done all this in a 20 cent bag..... sigh.....

The hood is going to make your life way easier. I use a rubber made container turned on its side at home. My contamination in the rubber made container is about 30 percent. At work I use a flow hood and my contamination is 5 to 10 percent.

Yes, they said table sugar works. Just make sure you don't accidentally use artificial sweetener or syrups. Most of those won't work well or at all.

I tried getting the chemicals from Sigma-Aldrich but they won't sell to individuals or home users. So ended going to eBay and Phytotech labs. Also, as he states you can use normal table sugar for sucrose.

@@joekunedo ahh I haven't had opportunity to perform tissue culture yet as I've been waiting for my orchid seed to mature. Orchids require similar procedures, will be able to test in about a month i think.

Devonly to break the dormancy period, you should try soaking your plant/seed in a solution of gibberellin.

@@fortuneflux I worked a year in an orchid nursery with a lab for flasked stock. We used banana and potato as the main food in the agar. Worked well.

Yup, that's the first hint to tell this is just a demo. Once they pulled it out of the bag to see it better and show the cameras it was confirmed. Good point though: clean, tight/well fitting clothing is an overlooked lab standard.

And as I've been in the industry of broad scale mass production with a dozen staff chopping away, it was always, hairy arms and beards that produced the most contamination. Even with hexafoam, body hair just carries too many bugs.
