For anyone who is wondering the door at the end brings you too a secret area above the ending area that has "True Ending" spelled out in coins. It leads to the same end but gives you a Yoshi and forces you into the pit after you win killing Mario while the laughing sound effect goes off.

If I recall correctly, entering that key door at the end mounts you on a new Yoshi and after finishing the level trampolines fall and make you and Yoshi fall to your death.

To use your words, "The level reset you" at that one point, because you did get that last coin, which brought out the key. The checkpoints remember the coins, but not the keys, which is a flaw in game design by Nintendo, at least I think it is. To answer your question at the end, if you had managed to grab the key, stay alive, and then go through the locked door behind the goal tape, the door would have taken you up to the top of the level, where written out in coins it says, "True Ending" and you would have been able to finish the level on a Yoshi, despite killing the ones you did earlier in the level.
