Immune explains how antibody-mediated depletion of myeloid-hematopoietic stem cells in aged mice restores characteristic features of a more youthful immune system, including increasing common lymphocyte progenitors, naive T cells and B cells, while decreasing age-related markers of immune decline.

Wow. Nice job. Can't get over how well Cindy presented the study and Brianne tracked it all and came up with fascinating insights for followup studies.

This was really interesting, thank you! I'm a TWIV / Matters Microbial fan, but this is my first time watching Immune. 👍 The gent who is spending $2M/year obsessively trying to reverse his ageing (Bryan Johnson) admittedly creeps me out. It's nice that he's trying to be as data-driven and methodical as possible (and making all of his data public), but being a sample size of 1, I'm not sure how useful it is. I guess every little bit helps... I just worry it's reinforcing to his zillions of followers that being old is somehow bad.

It just seems very odd to me that whatever the different ratio between the two types of cells in older mice would be deemed 'unbalanced' necessarily. Isn't there a possibility that the different ratio in older mice actually serves a necessary purpose? I assume there are other age-related changes in mammalian biology than just the ones in the cells of the immune system, a lot is going on in aging. Maybe that different ratio is in fact helpful to older mice for other reasons than just the ones studied in this paper. I would want to look at overall longevity of the different experimental and control groups, not just at their immediate response to a viral challenge. That seems a little short sighted.

So based on this theory of naive and memory Tcells. Would it be beneficial to travel to remote locations and get foreign infections? For example I went to Egypt for 2 weeks and was sick each week. That same year I went to Europe for 3 months and was not sick once. Not even for a day.

On the thymus and immune system, Prof Janet Lord PhD has published on long term older exercisers (around 60 to 80 yo I think) and shown that their thymus ages at a much slower rate and parts of the immune system are decades younger. Dena Dubal's research, and others, have shown that exercise significantly elevates alpha Klotho (and platelet factor 4) which helps keep the brain younger and may stave off dementia. The Conboy lab has done work (2020 paper) in which 50% of plasma (not blood) was replaced with saline (and albumin) in old mice (no young blood) and found significant tissue regeneration. Limit testing in human using an analogues process (TPE) also seems to have benefits. More work needs to be done in the plasma exchange area but it seems simple and promising as we can easily donate plasma at our local blood bank. Valter Longo's lab has shown that short bought of fasting (see his work on fasting mimicking diet) shifts the lymphoid to myeloid ratio to more youthful state. Mean-time, eating well and exercise will go a long way to supporting immunity.

Good discussion. Always have concerns about unintended consequences that aren’t seen in the short term. TY all for suggesting things that might be looked at in future studies.
