What to do with your brand new TWSBI ECO fountain pen? What are the tools for?

The pen already comes pregreased from the factory, just clean, ink, and go

Is there ever a concern that the piston will rotate inadvertently? Like when picking it up, handling it in your bag, etc?

No it comes prelubed it's for longer term maintenance or if you want to fully disassemble when cleaning between different inks.
