although grafting tape seems to work, parafilm has a much better gas permeability (estimated 10-fold for oxygen and 5-fold for CO2). And even if your roll is old, with a bit of practice it still can be stretched without tearing, especially if you only need to stretch in one direction. I am still using my roll which I bought almost 20 years ago! The problem with the plates sticking together only occurs when you use strips for sealing that are too wide and wrap around the top and/or bottom of the plates. I seal each standard petri dish with a 0,5" x 2" (12,5 mm x 50 mm) piece of parafilm. This way the plates can breathe very well without drying out, the plates don't stick together and the cost for sealing one plate is below 1 cent.

Even after understanding all of this it's still so peaceful to watch the process. Mycology is so awesome.

I’ve found cutting a piece of agar and barely touching the print on the very edge and put it back on agar but I like that way to! Just let the sprite print dry before putting away

Loved your work with the agar but in the micro lab we always have the plates upside down for the reasons you mentioned. We plate and store them “upside down.”
@RealSimpleMushrooms7 ай бұрын
Good point! I'll only store mine upsidedown if condensation is present

@@RealSimpleMushrooms the only time we have the plate right side up is just to transfer colonies. Keep it upside down to look like a pro baby.

That's funny how you mentioned using a reciprocating saw to shake them up. I was thinking the same thing right before you said it because that's how I shake up old spray paint cans. I use a long blade and tape my can to it.

Great vid as always...thanks. I've got some serious workspace envy for your "Lab with a View", and looking at it makes me want to add a lab extension to the house. Really appreciate the methodical and professional approach to your videos. Thank you.

@@RealSimpleMushrooms Nice work. New subscriber. Appreciate the liberal alcohol use. Just up the road in Albany. Watched an old English dude David Barlow’s KZbin videos long time ago. Really clear easy straightforward technique with easy to find items. Since been taken down. videos run thru from spore to casing and fruit MEA with manure done in jelly jars instead of plates to brown rice and manure in qt mason jars. His technique was scrape off the merest speck of spores from the print and then prick the plate with the tip the scalpel. He said he could make a quarter size print last him years Seems to work consistently and may be easier Especially since you got that dope hood Can’t wait to watch more of your videos. Thanks

does the potato agar work the best for the other facets of agar usage, like cloning, or is it only best for spores?
@RealSimpleMushrooms2 ай бұрын
It works great for all culture work but I’ve found that the success rate for germinating spores to be much higher on this recipe than any other I’ve tried

I couldn't agree more when it comes to paraffin and grafting tape. Parafilm tape sucks and it's way overpriced

Great and informative, I'm new to this field and I'm learning every day, and I'm in the middle of my first grow these days. Is the fan working at very high speed? The sound seems like.. If it is, high velocity air flow disrupts laminar flow and makes vortex. Showing the results of your work, would increase views dramatically, will take time of course, but pays off in views number.
@RealSimpleMushrooms10 ай бұрын
No this hood is designed really well and provides a great steady flow. I have 2 pre-filters on this thing to extend the life of the HEPA filter so the fan does have to work a little harder. And it's an industrial Dayton blower, not really designed with quiet in mind lol. I did a follow up video of the germination of these plates. Go check it out. :)

For spores a simple potato/agar mix and for everything else I use a LME or Sorghum Agar. I will be doing a video soon on my recipes

Would definitely like to see how you used to do agar with a SAB, I currently use a SAB that is a big tub I just hang over the counter and work in it from underneath. Saving up for a flow hood. Is that the 12x12 or the 24x18?

I might do one down the road. This is the 24x24 Bonsai.
