Does it compulsory to assign a licence to mail users while creating it. As in my organisation there are more than 1000 plus external vendor whose mail users need to create. Does I can also add a external routing address to communicate vise versa

You can create users without license for that purpose. Mail users dont require licenses in Exchange online.
