I should How to play your music off a USB Flash Drive, instead of playing it off a CD.
NOTE: Just be sure its in a MP4 Format

When i connect the lightning cable for iPhone it doesn’t show on USB 1 but its charging, and the music isn’t playing. Do you know how to fix it ?

I don't know this is just how to use a USB Device to listen to on the Ford Escape, not the iPhone so I don't know. You'll probably have to use the Bluetooth

Whan you play music from the flash drive, can you play a set in "random" mode?

You can have it play where it's got all the songs alphabetically one by one, but I don't know about having them all play randomly.
