Great to see people practicing in a SAB. When I see people with laminar Flow Hoods, I know I can't get away with the same things they are doing.

Agreed. As a beginner using SAB it's hard to learn proper sterile technique when it comes to working with agar because most videos have fancy flow hoods. e.g. it's one thing to read about how to streak a plate on some forum, but it's much more valuable to observe each action within a SAB--however videos within SABs are few.

Just think of the air inside the box/around as water. Practicing with SAB is much more budget friendly for amateur mycologists. Flow hoods make things hell of a lot easier though

A flow hood can be built for very cheap. Under 75$ and it'll last your entire life. The only thing you need to be wary of is covering the filter when not in use. But dousing things in iso doesn't sterilize anything.

Great video. Im just curious about one thing tho. Why did you put the agar wedges in the jar in one piece. Wouldn't cutting it up in multiple small pieces give you more innoculation points for faster growth? Idk, so I'm genuinely curious.

what do you do with the jars after you get them out of the pressure cooker? I've been using bags, I fold the openings underneath so that the bags sit on top of them. And I let them cool down overnight in a tub that I sanitize. However I have to then take them out and transfer them into the SAB, and I feel like these two transfers are where my contamination is likely coming from. Any advice?

yes, unless you are using the plastic lids, in which case you will need to just slightly unscrew.

@@linusgoblin Soap is far superior to bleach as soap is much better at trapping contaminants. That’s why we use soap to clean our hands, it doesn’t kill the bad stuff but it sticks to them and washes away when we rinse with water. The SAB is not meant to literally be sterile inherently, so we’re not trying to kill all contaminants but simply get them out of the air inside the SAB. Not only is bleach less effective for this purpose, it’s also not healthy for you or the environment, plus it will damage the plastic over time.

Very slightly, although that tiny amount is not all that big a deal, as you will be moving your hands in and out to sterilize and sanitize anyways, bringing in SOME contaminants. The point is to keep the air still when you’re working in it more than anything, you can have good success working in a porta potty with good sterile TEK.

You can also use a disposable coverall and just keep it in a clean place. It's probably even better because it'll hold your contamination in.

Why not spray soap water before you put everything in? Trying to cover your jars and stuff with foil seems counter productive and adding extra unnecessary steps.

Because that would be counter productive as the point of the soap water is to settle all the contaminants on the bottom of the SAB. If I bring things in after spraying, then now I’m bringing in outside contaminants into the SAB that are not settled on the bottom of the SAB.
