This guy is my hero...100% got me through undergrad science classes. I joke that I went to the University of KZbin because excellent online content like this is so much more helpful than many of the lectures and TA's were. Such a talented teacher. I love that he doesn't dumb it down, isn't afraid of detail, but breaks it down into digestible pieces that make sense.

I think it is a nice question, but remember we are talking about enzymes inhibitor, so I assume that it will hold it tightly that it won't be able to release the product.good luck

rahimah ismail it has to do with the inhibitor sequestering enzyme bound substrate complex (ES). When the inhibitor binds to ES the substrate is unable to leave the complex as a product, as a result it gives an apparent increase in affinity of the enzyme for the substrate. That's why Km decreases (it indicates that apparent increase in affinity). Hope this helps ;)

for mixed-noncompetitive inhibition, I think both Km and vmax are changed. This is different from the one you talked about in your video, which is, more precisely, a pure-noncompetitive inhibition.

Fantastic video - thank you! Question though: how does catalytic efficiency change for each of the 3 reversible inhibitors? It would make sense for it to decrease (since they are inhibitors), but I have a question. Since catalytic efficiency is defined as Kcat/Km, I would assume that a competitive inhibitor would decrease catalytic efficiency because Km increases and Kcat is the same (since Kcat = Vmax/[total enzyme] and Vmax & [total enzyme] don't change). But for an uncompetitive inhibitor, both Km AND Kcat decrease. Wouldn't this not change the ratio of catalytic efficiency?
