you say at 3:30 that cells with highest capacity of growth predominate - does that mean as you create more subcultures, you will get more and more of a single cell type (being the cell type with highest growth rate)?

if we make cultured for endothelial cells these cells may not accurately reflect organ specific endothelial cell characteristics what is the reason ? ist related to aberrant in gene expression or what happen ?

Very nice question , in reality extracellular matrix plays important role 8n tissue morphogenesis and cell fate specification . If we use isolated cultures, all those external signaling will be absent and hence fate specification could be altered . It's always a problem for cultures

Generally after 7-10 passage it should not be used for experiments....cells might accumulate many mutations and undergo telomere shortening...but this is highly variable for different cell lines
