Can anybody explain, how floxed mice used to be generated before CRISPR? People usually say that Cre mouse and floxed mouse can be generated by CRISPR/Cas but Cre-loxP system is older than CRISPR.

I have a question about the "CRE-DNA intermediate I" in the slide of "recombination mechanism", you noted the phosphate group and hydroxyl group, but isn't the phosphate group at 5'end, while the hydroxyl group at 3'end?
