What's the difference between CRISPR Cas9 and Cpf1?
➜ Dear fellow biotech lovers, thanks for watching our first episode of Molecular Minutes! In this episode, we introduce you to Cpf1, a new CRISPR nuclease discovered by the Zhang lab which we'll soon make available over here at abm. Cpf1 is smaller than Cas9 and targets complementary areas of the genome, making it an excellent addition to the CRISPR dream team. I do think that Cpf1 has a lot of potential because of its sticky end cuts and that it doesn't need cRNA. However, because Cpf1 is pretty new, while Cas9 has already been around for years, so perhaps people don't want to use Cpf1 over Cas9 for now since that would mean a lot more research and trials. But who knows, I hope Cpf1 gets more interest in future.

CRISPR Cas9 is in many late stage studiesand if and when approved their technology will get an immense boost. There will always be newcomers but the true and tried can not be easily swept aside.

can somebody explain between T-rich PAM sequences and G-rich PAM sequences?

Hi Rakib, thanks for watching and commenting -- Cpf1 is considered to have a "T-rich" PAM sequence as it's PAM sequence is "TTN" whereas Cas9 is considered to have a "G-rich" PAM sequence as its PAM sequence is "NGG"! Hope this helps!

Hi Rakib. I have a better explanation. T-rich PAM sequence is T-rich and G-rich PAM sequence is G-rich. Hope this helps!
