What I like about Black Widow is she isn't just an assassin but she's also a super spy which makes her more dangerous. Both Black Widow and Lady Shiva are excellent fighters but Natasha received a Red Room's version of Super-Soldier Serum that enhanced her immune system. It makes her physically stronger, immune to most diseases and physical disorder. She's also an expert marksman and mastery of various weapons and don't forget she also uses a variety of advanced equipment to help her fight enemies. She had been trained since she was a kid and she has been in many fights for so long. She also completed a lot of missions which makes her an experienced agent. There's a lot of things about Black Widow which makes her a better assassin and I think that's why they put Black Widow higher than Lady Shiva on the list

I am surprised that Raven is not featured. Her mutant ability to shapeshift to look and sound exactly like anyone else puts her at the top of the list. She is an exceptional and formidable martial artist. Though her fight skills and use of weapons are not on the level as some others mentioned here; her unique mutant ability enables her to topple governments and organizations all on her own without blowing her cover. It means no target, no matter how high profile, powerful and well protected he or she is, has no defense against Raven. It also means if Raven's assignment is to kill each of those mentioned her, even if they got the intel beforehand that they are her targets, none of them can do anything about it. Only await their fate.
