The movie never sets a seen where Wolverine learns that Deadpool has insane healing capabilities. I like to think that Wolverine stabbed Deadpool thinking he would die. Once Deadpool immediately stood up, Wolverine wasn't surprised and didn't care because he had seen so much in his lifetime.
