It means the writers and producers of the show intended it to be a certain way. An event or relationship that is obviously stated and alluded to directly within the show. Something that isn't just merely speculation. What they are saying is that for years on this show the subtext for a possible relationship between these two characters have only been hinted at, "queerbaited" and the fandom is divided by people who think they are and people who think they aren't meant to develop as a couple. There has never been clear cut evidence of a romantic feeling or an event that is clearly meant to be seen as a declaration of love until the scene in 15x18, which was Castiel's final scene on the show. The clear and blunt " I love you" and the statement about " wanting something he knew he couldn't have" has most of the fan base confident that this was, in fact a romantic love confession from Castiel to Dean, whether or not Dean reciprocates these feelings are still under speculation. Hope this helped you understand better if you aren't familiar with the online nature of the fan base.

Let me try it (English is not my first language either). When something is *Canon* it means that forms part of the original story and continuity. For example: Some comic books/events (Marvel and DC) occur in alternate universes, even thought those stories are happening do not form part of the "main story" or "main continuity", those "not canon" stories occurred by their own side without affecting or having any repercussions on the original continuity. Now, Destiel has always been a fan theory but it was never confirmed by the show until the episode 15x18. Once they confirmed that Castiel was actually in love with Dean that means that Destiel (non canon) it's not just a theory anymore, it's canon (part of the main story and original continuity). I really suck at explaining, but I did try.

I love Sam and Dean's relationship, the relationship and chemistry that has kept the series on screens for 15 years. SPN is over and I don't think we'll see such a brotherly love relationship on the screen right away. Dean is a lover of women and he is heterosexual, terefore he could never return love for the same sex. That's why Dean's almost terrified reactions. If someone of the same sex unpacks love for me, I run away and I don't want to see him. Jensen played it very right. Casse and Dean's relationship has always been complicated and full of violence and lies. So a friend - certainly, even if friends don't treat each other that way either. Love - never by Dean. So does the canon mean the expression of one side? Probably not.
