ma'am thank you very very much... this was the most helpful vdo for me in 2018... keep making these cool stuff.. coz only we the students know how it feels when we dont understand anything...cramming remains the sole option but it is disastrous in the long term... 2 minutes ago i was getting frustated but i am so happy and calm ☺😏

It is to see the gram NEGATIVE bacteria better. Since they won’t retain the crystal violet following destaining, you need a counter stain to make them visible.

If you accidentally used saffrinin instead of crystal violet but then did all the rest of the steps would they all just look pink? Or does the thick cell wall only bind to the crystal violet?

What will happen if you used the decolorizing solution first instead of the primary stain?

If you swap the order, the decolorizing solution won’t remove the crystal violet from the Gram-negative cells, and the saffranin will not be visible. All cells (Gram-pos and Gram-neg) will be purple.

Decolorizing is practically the most important step (obviously, all are necessary) because the decolorizing step is where you get the differences between cell types. After the crystal violet and iodine, ALL cells are purple. The alcohol decolorizing solution removes the purple from Gram-negative cells, but not Gram-positive cells (unless it’s on too long). Then saffranin is used as the counter stain to make the Gram-negative cells visible so you can see them under a microscope. The saffranin would not show up on the Gram-negative cells if they were still purple because you had skipped decolorizing.

shouldn't cocci(circle shaped) be the violet one, while the bacilli (rod shaped) be the pink one?

No. The cells in my video are generic examples. You are thinking of E. coli as a pink bacillus and S. aureus as a purple coccus because these are common examples. But there are many Gram-negative (pink) cocci like species of Neisseria (diplococcus) and Haemophilus (coccobacillus). And there are many Gram-positive (purple) bacilli like species of Listeria and Clostridium (both rods). In other words, cocci and bacilli can stain either color, depending on the species.

What happens if you forget the mordant or decolorizer for gram positive and gram negative?

It you forget the mordant, the crystal violet dye won’t stick during the decolorization and everything will end up pink from the safranin. If you forget the decolorizer, everything will end up purple because the crystal violet will be retained in both cell types.

Technically, BOTH are required because it is a differential stain...one is used to show Gram-positive bacteria and one is used to show Gram-negative bacteria. If you want to get picky, you might be able to argue that the crystal violet is the main stain and the safranin is just the counter stain.

The name Safranin really sounds blue-purple because of sapphire blue stone. And the crystal violet just sounds like some pink illicit pills. So, this thing is really messing up our brain.
