Therefore, what is described in books, when the nutrients in a liquid culture are depleted and the cells enter a state of anabiosis, I think is partly true. Also, a very interesting experiment turned out with a syringe that was stored for more than 2 years at room temperature. The Kuehneromyces mutabilis culture, when inoculated onto a new liquid medium, showed phenomenal growth, unlike the mother culture I purchased, which did not grow as actively. This is to say that not everything has been studied and described about liquid cultures. You have my like and subscription, I watch your videos. Greetings from Ukraine👋

Very good information! Thank you for sharing 🙏 There is much to be learned about liquid cultures. It is very fascinating the difference between species and their reactions to certain nutrients or conditions.

I also performed this manipulation a few months ago. The jars are now in their third year, and I believe it's a wonderful way to get rid of excess fluid and metabolites. I also decided to add a sterile nutrient medium to the jars, but with different ingredients, which gave a boost and revived the culture

That’s awesome! That was actually my next experiment, removing the old liquid and adding new nutrient liquid to see if it continues growing. Thanks for watching! 🍄

@@SporenSprout I've concluded that there's a high risk of contaminating the culture when transferring new liquid into the old, even though the contamination isn't visible. But this transferring becomes tiresome if it's not just one jar. It's much easier to create a new liquid culture. Now I just be draining the excess liquid, and the culture that's left will be transferred into 20ml syringes and put them in storage, to prevent "JAR-Apocalypse🤣Which quickly occurs when many strains and cultures appear

Thanks for the video. It's always good to see other people's techniques. I find that the more LC I use from and old Jar to start the new one, the quicker and stronger it grows. This is especially noticeable when using an LC recipe with peptone, LME and honey. I thought one of my Pioppino jars had contamination, but on closer inspection it had filled out about 95% of the broth.

What are the signs of mold contamination in LC jars? I've seen bacterial before but how can I tell the difference between mold growth and mycelium in LC?

Any use of the liquid you removed from the jar? 😊 i like to use everything i can! Also thanks for your agar sandwich video! Works like a charm!

I think a peristaltic pump, tubing and luer lock to tubing adapters would be a good machine. I'm trying it for $32 on amazon.

In a cool basement, what should one expect for maximum time to grow ? I've only got one jar with significant growth in it after about a month.
@SporenSprout7 ай бұрын
3 months is good

So I should see healthy growth within 3 months ? Or do you mean they expire after 3 months ? Thanks@@SporenSprout
@SporenSprout7 ай бұрын
@@SmeeUncleJoe They should have healthy growth in 1-3 months. They won’t expire for 6-12 months or longer

@@SporenSprout was your LC made with honey. Or corn syrup? One is asking as my bears head solution is a light yellow in colour, not cloudy. I used corn syrup LME with peptone. The mycelium look white. I will test it on agar to see if it’s contaminated. This is my first time making liquid cultures. Is this ok or is there a chance this could be contaminated ? 👍

@@SporenSprout I saw that video and it was great. Now I'm wondering if we should grow the mushroom inside the plate or can we plant Spanish seeds inside the glass.

In the video, when you put the tissue on the plate, what dye did you inject on it?
