During equipment running can I take a fresh back up from PLC ?? What all care I need to take ??

Yes you can take the fresh backup from running system ,keep in mind before connecting laptop to machine network check you laptop IP it should not conflict with any other device in machine network and select correct version and carefully choose upload option
