It is (or was, hard to stay updated with comic canonicity whey they hit the reset button so often) canon, in civil war, after Peter sided with Tony and revealed his identity, KP hired a hitman to kill him, but he missed and shot aunt May instead (this eventually led to One more day, but this is before that) , needless to say, Peter was pissed, so he went AS PETER PARKER and beat the absolute $#1+ out of him, and then he straight up told him he was gonna kill him, just not that day. This short would basically be Peter announcing he came to finish the job.

For those who don't know this is a reference to the comics, in the comics kingpin attempts to have Peter assassinated but the person who gets shot instead is Aunt May Peter then goes to the prison that is holding kingpin removes his costume and proceeds to absolutely annihilate him, he then tells kingpin that if Aunt May doesn't pull through he's going to come back and kill him as Peter Parker.

This is based on a storyline in which the whole world already knew his identity because he’d revealed it during the Civil War comic. Fisk put a hit out on Peter Parker and the assassin missed and shot Aunt May instead, hospitalizing her. Peter wanted him to know it was Peter Parker coming for Fisk because he didn’t attack Spider-Man and it wasn’t Spider-Man coming to get him. The very knowledge that made him vulnerable to Kingpin is what endangered Kingpin in return. In the end, he threatened to kill Fisk if Aunt May died, but he wound up making a deal with Mephisto to save her, anyway (at the cost of his marriage), and restored his secret identity in the process. Fisk is still around and probably doesn’t even remember any of it (and if he does, he doesn’t remember why Spider-Man beat the him senseless).

For anyone curious: kingpin put a hit on Peter’s wife, Mary Jane, and the shooter accidentally hit aunt May instead. Peter figured out it was kingpin and paid him a visit in the middle of prison, literally. Put kingpin on notice in front of all of GP, telling him that he was going to be back (one day/week, I forget), and when he returned they were gonna fight… It was not a fight. Peter beat kingpin, badly. Before threatening every other inmate, that out on the street, him stopping them is just business, but going after his family: you’re looking to catch a body.

In this specific context, it's based on a storyline where his secret identity leaked. Kingpin put a hit out on Aunt May and out her in the hospital. Peter then worked his way up through the henchmen to find out who did it. He eventually found out it was Kingpin. When he found out it was Kingpin, he immediately went to the prison he was being held at and broke in. Where he proceeded to take off his costume and confront him as Peter Parker rather than Spiderman. At which point he went to work beating the absolute hell out of Kingpin in the middle of the cell block. There were no webs slung, no clever quips said by spiderman. Nothing but hands being thrown, as Peter decided to not use the restraint he usually does. He beat Kingpin to within an inch of death and warned him that killing him as as simple as shooting webs into his throught and letting him suffocate to death. He also warned everyone that this is what would happen if they touched his loved once, and on his way out he told Kingpin that he would be back if Aunt May died. Letting him know that if she died, he was a dead man walking.

For those who do not understand, i gotchu. Theres a version of spiderman who leaked his own identity cause of the marvel comics civil war, and as we all know the writers love to make peter SUFFER. So his aunt may gets shot and he finds out kingpin did it. So as you can guess, he was BEYOND PISSED, and went to the jail kingpin was at. He went there, and kingpin wasnt very scared. He announced he wasnt there as spiderman, the virtuous hero of nyc, but as peter parker. he then proceeded TO BEAT THE LIVING HELL OUTTA KINGPIN, NO HOLDS BARRED. he stopped holding back his punches, and then lifted his wrist to kingpin's mouth and said that he COULD DROWN HIM INTERNALLY WITH HIS WEBS IN A FEW SECONDS, TOLD KINGPIN NOT TO MESS WITH HIS FAMILY, and then left, kingpin was AT 1 HP. Kingpin was traumatized and embarrassed.
