I am a college student at the University of Texas in Austin and I am in the process of studying this for my test. I was reviewing your video and I found a mistake that can get all the people watching this video confused. The repressor doesnt no bind to the promoter. RNA polymerase only binds to the promoter. After the promoter in the sequence of DNA there is something called the operator, this is where the repressor and activator binds.

LacO1 is located within the promoter. But the activator can be upstream to the promoter. So, the video isn't incorrect but it could have been more precise.

the promoter is the binding site of RNA polymerase not of the repressor protein LacI. The repressor binds to the operator that is located after the promoter region.

As far as the upper division genetics course I'm taking required for my major in the biological sciences, this video went into just enough depth to cover everything I need to know.

The repressor binds to the operator, to the promoter. I'm not sure if this is an OCW thing meant for AP students or not, but at a university level, this video misses some crucial details. Still good for general concepts though.

I think your model has a big problem , as i know the promoter is a certain sequence which promote binding of RNA polymearse - after promoter there is separate place called : operator , the repressor is actually bind to the operator not directly to promoter . maybe i'm wrong but please explain it if possible thanks for your video ; )

i think that there is a little mistake out there , the repressor doesnt bind to the promoter the activator neither they bind to the operator .. the promoter only binds to the RNa polymerase , am i right ?

actually, allolactose binds to the Lac repressor, not lactose.

Incomplete explanation. RNA pol binds to the promoter. It is the repressor which binds the operator (which is not shown here).

this video is too basic. Hard to believe that MIT is teaching this kind of class to its students. They forgot a lot of details like the other comments are mentioning. but its good for a basic understanding i guess
