Ga exists in up to 16 isoforms, grouped into 4 subgroups. a(s), a(i), a(q) and a(12/13). If the receptor activates a G-Protein, that has an a(s) subunit, this subunit can activate membranebound adenylate cyclase, which produces cAMP. this is is a second messenger activating PKA. PKA is a serine/threonine-kinase, which phosphorylates different proteins and therefore activates other cascades. An a(i) subunit inhibits the the adenylate cyclase, which results in low cAMP levels and no activation of PKA. a(q) subunits activate phospholipase C. This enzyme can divide PIP2, a subtance in the cellmembrane into IP3 and DAG. the DAG is left in the membrane, the IP3 binds to calcium channels of intracellular calcium depots (e.g the ER). calcium concentration rises. Another enzyme called PKC binds to the DAG in the membrane. It also needs the calcium for activation. With both, DAG and Ca avaible, it acts as a serine/threonine kinase and can phosphorylate other protein, which results in activation of other steps. the a(t) subunit is a special form of an a(i) subunit. it is mostly expressed in rod cells of the retina (T stands for transducin, a synonyme for this kind of g-protein).
