U can used a presto pan as well, in fact u can add aftermarket preasure meter in stock pan

Why do you adjust ph before you add the gelling agent? Doesn't the gel affect the ph?

Most labs do it this way, a professional grade gelling agents does not chance the pH.

Gelling agents like agar also won’t change pH.

How many watts is your microwave?

A pressure cooker/autoclave at 15psi sterilises at 121deg (run for 15-20mins once full pressure is reached) .... 21deg above atmospheric pressure.... Microwaves can only raise to 100deg. regardless of how long or at what power it is run for, so no better than a kettle or a saucepan. The microwave method can only reach 100deg since it is not pressurised so what other factors do you think contribute to producing sterility of media when using a microwave.... how reliable do you find microwave use?

It's very reliable after you find the right amount of time and power. Not all microwaves have the same power even if they use the same wattage. For my microwave, ten minutes at 50% power is good for 16 oz deli containers.

Do you have some special plastic cups? I tried in a microwave at a similar percentage and they melted about 3 minutes in. I did not leave the lids on tight, they were loose. Does the mixture have to start boiling?

Make sure they are made out polypropylene (PP). Most lids are made out of PET, which cannot stand the high temperatures.

@@PlantCellTechnology What brand/supplier do you use to obtain your 4 oz PP cups and lids? I have searched everywhere I can and can only find ones with PET lids. Any guidance would be appreciated.

Is it necessary to clean or sterilize the microwave itself before using for sterilisation?

You can do it for extra precautions in you process. Otherwise you can wipe and clean the inside surface and sterilize the media.

You can. But, it's not recommended due to the high risk of contamination. You can create a culture area using a plastic box and keeping an air filter closer to it.

@@agrippanyagwaya2308 You can check our videos in which Francisco is teaching about building labs. However, you can just place the air filter near the culture area of yours. Ther's no trick involved.

Hey! I appreciate your content videos. I am curious if the PP containers can be autoclaved for ms sterilization (121 degrees Celsius)

Polypropylene containers are autoclavable. The recommended autoclave cycle for empty containers is 121°C at 15 psi for 20 minutes. However, care must be taken to allow free air circulation into and out of vessels during the autoclave cycle, especially during the venting and cooling stages.

Thank you Sir, im new subscriber, im interesting in making plants/ tissue culture. So for starting Sir can use electric oven? Because autoclave is so expensive.

You can use a pressure cooker with a pressure gauge. It should be cheaper. Ovens can be used but they aren't effective and not suitable to sterilize every equipment/tool.
