When putting a screw back into the same hole in wood, it's best to use a manual screwdriver. Put the screw back in the hole, go in one full turn, but then back it out a quarter turn, before continuing to drive it all the way home. This prevents the screw cutting a second thread in the hole, and potentially stripping it out. If the hole is accidentally cross threaded or stripped out, cut a match in half with a knife, feather out one end of that, and put it in the hole with the feathered end on the outside of the hole. I've used the match trick to rehang doors where the hinge holes have been stripped out, it's strong.

What I did and worked out fantastic was to buy the thick clear plastic cases. Then I drill out the small holes so I can use a #6 screw. Since doing this, I have never had a mezzusa come off of the wall. To understand the context of why I need such a strong attachment, the mezzusas are in a commercial building.

We use Velcro Heavy-Duty Self Adhesive to put up our Mezuzahs. Less messy!

Hypothetical question, if someone is having a house built, can a craftsman build a holder into a custom door frame or does it have to be a separate holder that gets attached?

It can be in the doorframe, in fact I believe that's how it was done in many instances before modern cases were "invented" I've put them in doorframes in factories where there is a fear of them getting knocked off. You just need the make sure they are not deeper then a tefach which is 3/4 inches.
