I have much to memorize for my exams and I've been feeling really dumb lately because I could not for the life of me understand the steps, but these visuals along with the simple description was AMAZING. I know that proteins are produced by the ribosomes, and that the ribosomes reside in the cytoplasm, not within the nucleus. So I can imagine that a protein that has been produced by the ribosome, but that belongs in the nucleus, would have to be transported INTO the nucleus. What I'm not clear on what kind of protein would have to be transported out of the nucleus. I imagine that any protein in the nucleus is supposed to stay there, since all proteins are produced in the cytoplasm and wouldn't be moved to the nucleus unless needed there.

Assuming NTF2 has interactions with NPC proteins that allows it to slide into only nucleus side and bring RanGDP back. Then another protein is supposed to bring NTF2 back that is only able to pass to cytoplasm side. And another protein is gonna have bring that protein back to nucleus and so on. Where does this loop end? lol

NTF2 binds RanGDP in the cytoplasm and then ferries it into the nucleus by virtue of the fact that NTF2 can also bind FG repeats on nucleoporins similarly to NTRs (Bayliss et al., 1999). NTF2 has a much lower affinity for RanGTP than RanGDP, and thus travels empty back to the cytoplasm after its cargo (RanGDP) is converted to RanGTP by RCC1 inside the nucleus triggering its release.

doesn't RAN-GDP bind Bêta-importin BEFORE the importation and is then exchanged in the nucleus by GEF ? I'm confused now
