I currently cultivate mushrooms and am fascinated by PTC. I had heard of it before but never figured to look it up until I saw one of your videos. Just wanted to add a quick note about pressure cookers; I have the same one but made for induction heating and I use it on a standalone electric stove. I start it at 1600w and wait until the safety plug (also check if the two halves are properly screwed together before running the cooker) pops up and closes on it's own, by that time you will have a steady stream of steam coming from the rocker spout, then I place the rocker on top and monitor it until the pressure builds up to 15psi. Then I lower the power down to 1200w and leave it there for however long I am running the cooker. No further adjustments needed. (on my specific stove, your mileage may vary) Always remember to keep an eye on it though as you stated. Your videos are super clear and nice to follow :)

I don't like the instant pot for autoclaving. It only goes to 12PSI, and they are excruciatingly tiny. Electric stove is fine, just use "medium high." The warning in the manual refers to the propane "jet burners" as opposed to household cooktops. My favorite is the Presto "induction compatible" model, gets to pressure super fast on a 3500W induction burner. I've never found any benefit to venting off steam for ten minutes. I believe that tidbit in the manual (and from the USDA apparently) is based on flawed folk lore. With or without venting, I read identical temperatures at 15PSI. Just my two cents from the field ;)

I originally purchased my current multicooker to use as an autoclave like 11 years ago. It was used to do some citizen science with animal cell cultures, which require significantly more sterile conditions than plants do. I didn't even have a laminar flow hood or a CO2 capable incubator and still got virtually no contamination.

Really good explanation! As someone who is interested into getting in plant tissue culture just for the fun of learning, may I ask, what container materials would you suggest kind of to compare the pros and cons. I know glass is 100% autoclavable but some people use plastic containers (I guess it would be cheaper) and I'm wondering what type of polymers are they made from? Also, can you use tight-seal mason glass jars in autoclaving or would the rubber melt?

Hi! You can use deli containers instead of glass jars if both the plastic container and the plastic lid are autoclavable (aka can withstand 121 degrees Celcius). I have never used a mason jar for tissue culture, but if you buy mason jars that are designed for pressure canning, they should be able to withstand the pressure cooker. If you're going to try tissue culture with mason jars, I would probably opt for mason jars that don't contain a rubber seal, like Ball mason jars

No. Water bath canning does not eliminate all pathogens.
