I understood that the differance between the restriction enzymes is exonucleases and endonucleases, but is there a specific target sequence for each enzyme and what is reason for this? is there a specific number of nucleotides for each enzyme?

I have a question, though. 3:16 you mentioned that staggered cuts are preferred in recombinant DNA technology as they will form hydrogen bonds easily (between strands) as opposed to blunt ends that'll need peptide bonds (within the strand/between nucleotides)?
