Rocket Immuno Electrophoresis

RIEP known as electro immuno diffusion is a simple, quick and reproducible method for determining the concentration of antigen in an unknown sample. Various concentrations of antigens are loaded. On electrophoresis, the antigen begins to migrate towards anode. Antigen and antibody interacts, when it reaches the equivalence point, Ag-Ab complex precipitates. The precipitation line is seen in the form of a rocket.

when they melt in the microwave it usually just gets in a liquid state, and they when it cools I guess they add the Antibody. And I think they also use a tampon on the antigen and PBS.

Good video! Thanks a lot. But I wonder whether the antibody would get denatured while put into the gel because the gel is usually pretty hot, right? Is there any other treatment to prevent this?

姚俊辰 at 50-60°C the proteins antibodies won't denature. So we should allow the gel to cool down to this temperature and then antibody could be added.

you really need to update yourself. we don't need to make wells manually. we can use a comb which is available in the market to make gel wells with appropriate size
