Lol @everyone reading waaaay more into this than it actually is.... The character Stefon has a thing for the other one and had unsuccessfully tried to 'do something' about it before and made off-color comments before. Finally got to give him a kiss (probably scripted as a short smooch that the actor turned into a bigger deal). No closeted anything, nothing between the lines. Just the actors having fun with some sexual tension between their characters. That's why the smile--it was a running gag (reflex) and as another commenter put it, Stefon's actor "won this round". That's all.

He didn't come up with a lot of 'this stuff'... one of the reasons why Hader and Meyers used to get the giggles so much during the Stefon skits is that writer John Mulaney would add jokes in that Hader would only see as he was reading it off the teleprompter in the live show.

What is so special about their love is the way that Seth, despite becoming reasonably exasperated with Stefon and realising that he's incompetent, still always takes time to praise his efforts and tell him he's doing a good job anyway. That's the ideal kind of partner support.
