this column chromatography lecture video explains about the protein gel filtration chromatography technique principle. This is also known as molecular exclusion chromatography or gel filtration chromatography. In this chromatography process molecules are separated based on their size. Mixture of molecules are passed through a polymer network made with agarose gel. Large molecules will not enter into the pores created by the agarose polymer so they will elute faster and the smaller molecules will be trapped inside the agarose polymer as a result they will take more time to come out of the column.

Sir What if smaller molecules enter the void along with larger.. its not compulsory that smaller take the pores only, they may enter the voids too. Than first to be eluted is smaller particles in this case..

sir , when we will pour a sample it may contains undesirable materials but we get product according to size. So undesirable materials are also disributed in product according to size. Then how we can separate undesirable materials.

Another difference between gel electrophoresis and size exclusion chromatography is that smaller fragments will run faster and larger ones will run slowly in gel electrophoresis while in size exclusion chromatography smaller molecules will come slowly and larger ones come faster, am I right Sir?

If I'm using agarose instead of sephadex beads as the matrix of the column then how do the protein of high mol. wt. will elute faster than that of lower ones

If sample contain both smaller and larger prtn then how only larger prtns I'll come through pores why can't small prtns come through the same pores from which larger one comes

In another lecture... they said larger molecules will travel slower as they want to find alternative way, around the st phase and but small molecules can easily move through the pores in them and travels faster.... Actually which statement is true ??🤔🤔

Aravind SR I think the one where large mol. Are eluted first is much more gripping concept because as you see the surface are for the large molecule will be greater and the way they had to find will be small in comparison to the small molecule which are stuck in the labyrinth of pores.....like a ant which has average speed way much faster than a cat but the cat will still outrun them.
